Application Abreviews Notes
IHC-P Use a concentration of 4 - 8 µg/ml. Perform heat mediated antigen retrieval via the microwave method before commencing with IHC staining protocol.
ICC/IF 1/250. Fix with 3% paraformaldehyde/2% sucrose (see review by R Drapkin).
WB Use a concentration of 1 - 2.5 µg/ml. Detects a band of approximately 34, 41 kDa (predicted molecular weight: 44 , 42 kDa).Can be blocked with Human Pax2 peptide (ab111670). Dilute antibody in 5% milk in PBS buffer. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
ChIP Use at an assay dependent concentration.

    Pax2 is a transcription factor critically required during the development of the nervous and excretory systems, including the midbrain, hindbrain, spinal cord, eye, ear and urogenital tract. Like other products of the Pax gene family, Pax2 encodes a conserved 128 amino acid paired box DNA-binding domain in the N-terminal portion of the molecule. Function: Probable transcription factor that may have a role in kidney cell differentiation. Has a critical role in the development of the urogenital tract, the eyes, and the CNS. Tissue specificity: Expressed in primitive cells of the kidney, ureter, eye, ear and central nervous system.

    why is the actual band size different from the predicted?

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human kidney tissue labelling Pax2 with ab23799 at a concentration of 4-8µg/ml. The arrows indicate the positively labelled cells, epithelial cells of the renal tube.

  • The human ovarian carcinoma cell line OVCA429 was used with ab23799 in ICC/IF. The cells were fixed in paraformaldehyde. Blocking was in 5% serum for 1 hour. The primary antibody was incubated at a dilution of 1/250 for one hour. Immunofluorescence (ICC/IF) only worked with 3% paraformaldehyde/2% sucrose fixation. When 70% methanol/30% acetone fixation was tried non-specific cytoplasmic staining was seen. In the left hand panel staining with DAPI is shown, and in the right staining with the ab23799 antibody.
