Application Abreviews Notes
WB Use at an assay dependent concentration. To detect hPDGF-AA by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hPDGF-AA is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
ELISA Use at an assay dependent concentration. To detect hPDGF-AA by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hPDGF-AA.
Neutralising Use at an assay dependent concentration. To yield one-half maximal inhibition [ND50] of the biological activity of hPDGF-AA (10.0 ng/ml), a concentration of 0.17 - 0.19 µg/ml of this antibody is required.
IHC-P Use a concentration of 1 µg/ml.

    Platelet-derived growth factor is a potent mitogen for cells of mesenchymal origin. Binding of this growth factor to its receptor elicits a variety of cellular responses. It is released by platelets upon wounding and plays an important role in stimulating adjacent cells to grow and thereby heals the wound.

All the information we have on species cross reactivity is specified on the datasheet. To our knowledge, cross reactivity with mouse has not yet been tested. The immunogen used was recombinant hPDGF-AA so I am unable to check the homolgy. You may be able to find more information on the sequence of the whole protein on Locus Link and Swiss prot (see the Abcam website resoucres section for more information). To our knowledge this antibody has yet to be tested in this application. We would welcome any feedback in this respect from researchers who have tried using this product. I would expect that this antibody would work in this application as it was raised against full length PDGF. The antibody has only been tested in human tissue, the reactivity with other species is not known.
