The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use a concentration of 1 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
WB Use at an assay dependent concentration.

This antibody has been tested in WB against purified recombinant protein only. Abcam does not have data on how this antibody performs in WB when using tissue/ cell line lysates.

Lane 2 : Recombinant human PDGF BB protein (ab9706)

Lysates/proteins at 0.01 µg per lane.

    All lanes : Goat Anti-Chicken IgY H&L (HRP) (ab6877) at 1/3000 dilution

    Developed using the ECL technique.

    Performed under reducing conditions.

    Observed band size: 16.7 kDa
why is the actual band size different from the predicted?

    Exposure time: 8 minutes

ab73231 has a His tag so shows a high MW.

  • IHC image of PDGF BB staining in Human normal kidney formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab139753, 1µg/ml, for 15 mins at room temperature. A Goat anti-Chicken biotinylated secondary antibody was used to detect the primary, and visualized using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
