      ab51869 at 1/200 dilution, staining PDGF BB in human placental tissue section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections).

      Overlay histogram showing SHSY-5Y cells stained with ab51869 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab51869, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in SHSY-5Y cells fixed with 80% methanol (5 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

    Can you please recommend an antibody to PDGF BB for IHC-P on human samples in mouse tissue? We are looking for an antibody that will show minimal reactivity with mouse and with PDGFA.

    Answer

    Thank you for contacting us, I apologize for the delay in my reply. I would recommend ab114130 for your purposes. The immunogen for this antibody shows no significant similarity to PDGFA, and it is slightly less likely to react with mouse than our other rabbit polyclonal antibodies. That being said, the mouse protein is fairly homologous with human and there is still a chance that ab114130 may show some reactivity to mouse PGDF BB.
    If you do want to use the mouse monoclonal antibody ab51869 on mouse tissue, you may need to perform an additional blocking step to prevent mouse-on-mouse background staining. If mouse-on-mouse background is a problem in your assay, it will be visible when running your no primary antibody or isotype control. To reduce this background, you can perform an additional blocking step after your normal serum block and before the primary antibody incubation. The sections should be incubated for 1 hour with a F(ab) fragment unconjugated secondary antibody to mouse IgG, such as ab6668, at a ˜1:100 dilution at room temperature.
