Target

  • Function
Receptor that binds both PDGFA and PDGFB and has a tyrosine-protein kinase activity.
  • Tissue specificity
Expressed in primary and metastatic colon tumors and in normal colon tissue. Tumors may express a different isoform to that found in normal tissue.
  • Involvement in disease
Note=A chromosomal aberration involving PDGFRA is found in some cases of hypereosinophilic syndrome. Interstitial chromosomal deletion del(4)(q12q12) causes the fusion of FIP1L1 and PDGFRA (FIP1L1-PDGFRA).
  • Sequence similarities
Belongs to the protein kinase superfamily. Tyr protein kinase family. CSF-1/PDGF receptor subfamily.
    Contains 5 Ig-like C2-type (immunoglobulin-like) domains.
    Contains 1 protein kinase domain.
  • Cellular localization

Images

  • ab15501 staining PDGFR alpha in human ovarian carcinoma by Immunohistochemistry (FFPE-sections).

  • Immunohistochemical analysis of frozen mouse embryonic submandibular gland tissue, staining PDGFR alpha with ab15501.

Sections were blocked with 1% BSA before incubation with primary antibody. An Alexa Fluor®488-conjugated anti rabbit IgG was used as the secondary antibody.

Answer

As far as we are aware, there are no published references that specifically feature these two antibodies. However, there are super images on each of the datasheets showing that the antibodies work well in immunohistochemistry and information is provided on how best to prepare formalin fixed tissues.

Answer

Thank you for your phone call. Unfortunately there is not an image available showing the skin testing. Both Human skin and Ovarian Carcinoma are recommended as positive controls for ab15501 and there is an image showing the staining with the ovarian carcinoma sample on the online datasheet. As ab15501 is in the pre-diluted form, and there should be no need to dilute it any further. With your brain tissue samples, I would use the antibody neat and make sure to boil the tissue sections in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min. If you still see high background, try increasing the blocking incubation period and consider changing blocking agent. If you need additional assistance, please do let us know.

Answer

I am sorry to hear you are having problems with ab15501. I would like to suggest an antigen retrieval step of boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at room temperature for 20 min as recommended on the datasheet. The source of the antibody was able to inform us that the predilution solution contains green food colour. Have you tried other prediluted antibodies and if so have you always got similar non-specific staining? If that is the case, it is probably because the lucifer yellow reacts with the green colour in the predilution solution. We will hopefully be adding to our catalogue the concentrate format of the antibody and you may wish to purchase it and dilute the antibody with 1% BSA in PBS to see whether the non-specific staining is gone. I hope this information helps, please do not hesitate to contact us again if you need further assistance,
