Target

  • Function

    Activates insulin, somatostatin, glucokinase, islet amyloid polypeptide and glucose transporter type 2 gene transcription. Particularly involved in glucose-dependent regulation of insulin gene transcription. Binds preferentially the DNA motif 5'-[CT]TAAT[TG]-3'. During development, specifies the early pancreatic epithelium, permitting its proliferation, branching and subsequent differentiation. At adult stage, required for maintaining the hormone-producing phenotype of the beta-cell.
  • Tissue specificity

    Duodenum and pancreas (Langerhans islet beta cells and small subsets of endocrine non-beta-cells, at low levels in acinar cells).
  • Involvement in disease

    Defects in PDX1 are a cause of pancreatic agenesis (PAC) [MIM:260370]. This autosomal recessive disorder is characterized by absence or hypoplasia of pancreas, leading to early-onset insulin-dependent diabetes mellitus. This was found in a frameshift mutation that produces a truncated protein and results in a second initiation that produces a second protein that act as a dominant negative mutant.
    Defects in PDX1 are a cause of non-insulin-dependent diabetes mellitus (NIDDM) [MIM:125853]; also known as diabetes mellitus type 2. NIDDM is characterized by an autosomal dominant mode of inheritance, onset during adulthood and insulin resistance.
    Defects in PDX1 are the cause of maturity-onset diabetes of the young type 4 (MODY4) [MIM:606392]; also symbolized MODY-4. MODY is a form of diabetes that is characterized by an autosomal dominant mode of inheritance, onset in childhood or early adulthood (usually before 25 years of age), a primary defect in insulin secretion and frequent insulin-independence at the beginning of the disease.

  • Domain

    The Antp-type hexapeptide mediates heterodimerization with PBX on a regulatory element of the somatostatin promoter.
    The homeodomain, which contains the nuclear localization signal, not only mediates DNA-binding, but also acts as a protein-protein interaction domain for TCF3(E47), NEUROD1 and HMG-I(Y).

    Dear support team,
    I am interested in an antibody against chicken Pdx1. Since currently
    there is none, I would like to know wether your Anti-PDX1 antibody
    (ab47267) crossreacts with the chicken protein. Could you tell me the
    sequence of the immunogen so that I can check how is the conservation
    of the used epitope between mouse and chick?

    Thank you very much for contacting us.

    We currently do not have the immunogen sequence of ab47267 available, but I have contacted the originator for the information you requested, and I am awaiting a reply. I will forward to you the sequence as soon as I receive it.

    In the meantime I have checked our other PDX1 antibodies for their potential cross-reactivity with chicken. According to my BLAST research on NCBI or SwissProt(http://blast.ncbi.nlm.nih.gov/Blast.cgi?CMD=Web&PAGE_TYPE=BlastHomeand http://www.uniprot.org/uniprot/), the immunogen sequence of ab94931 reveals a sequence similarity of 100% to the chicken orthologue, and thus should cross-react:

    Please note that this antibody is a fast-track antibody: These antibodies have undergone variable degrees of characterization. They have all been affinity purified and show high titre values against the immunizing peptide (by ELISA). Fast-track antibodiesare released quickly to meet customer needs and requirements; the products are still undergoing further tests and characterization steps. Therefore, we release Fast-Track antibodies at a reduced price. Please read the terms of use on the datasheet for further information.

    Also, I have checked the following antibodies:

    ab84987: 72% to SwissProt E1C2J1. Candidate for testing? Please contact us, as you might be eligible for our testing discount program (https://www.abcam.com/collaborationdiscount).

    ab19385: 60% sequence similarity. Unlikely cross-reactivity.

    ab98298 (immunogen AFQRGPAPEFSASP): 57% to chicken PDX1, but 100% to a number of other proteins, so cross-reactivity to PDX1 is unlikely, staining of other unrelated proteins likely. Therefore, not suitable for testing.
