      The H-H-H-H-H-H motif is used as a tag on many recombinant proteins to facilitate purification. The antibody recognizes the His-tag fused to the amino- or carboxy- termini of targeted proteins in transfected or transformed cells.

      BALB-3T3 cells, which express moderate amounts of EGFR, were incubated with a His6-tagged anti-EGFR fibronectin domain. The labeled cells were washed and incubated with 50 microliters of 1:15 diluted PE-conjugated anti-His6 antibody. Cells were washed and analyzed by flow cytometry.     We have purified recombinant protein containing polihystidine tag in a solution extracted from E.coli cells. We want to confirm that our solution indeed contains those proteins so that we will use it in our experiments. One of our collaborators has your

    and he offered his antibodies to us. However this antibody seems to be used for Flow Cytometry only. How can we use this antibody to show that our solution actually contain his-tagged proteins?

    Thank you for contacting us. Indeed we have only tested this antibody for Flow Cytometry. You may wish to try an ELISA to test your solution, we have a protocol for ELISA with a fluorophore conjugated primary antibody here:

    We cannot guarantee the antibody will work for this application however. We do have 6X His tag antibodies that have been tested for ELISA such as ab9108.

    Inquiry: Dear Sir/Madam I have a question about the antibodies that I am going to purchase. I am going to purchase a Anti-6X His tag (PE)antibody (ab72467) for my Flow cytometry analysis on bacterial cells (Salmonella Typhimurium). I just want to find the 1. Ab concentration of the product, 2. the F:P ratio, and 3. a suitible isotype control. I've narrow down the isotype control to two possible mAb (PE), which are (ab1280) and (ab91357). Is it possible for these isotype controls to react with Salmonella typhimurium proteins? are they raised to against any bacterial proteins? I's also like to know their Ab concentration and F:P ratio too. Thanks very much

    In answer to your questions:
    1. Ab concentration of anti-6X His tag (PE), ab72467
    The concentration of this antibody is 69 ug/mL.
    2. The F:P ratio of the antibody
    The F:P ratio of ab72467 is 1:1
    3. A suitable isotype control
    I think either of the antibodies you have suggested would be suitable as an isotype control. The first, ab1280, is supplied at a concentration of 23 ug/mL and the F:P ratio is lot dependent. The current lot is a ratio of 0.9. The second antibody, ab91357, is supplied at a concentration of 7.5 ug/mL and the F:P ratio is between 1-2. It is very unlikely that either of these isotype controls will cross react with your Salmonella typhimurium however as we have not tested this ourselves we would be able to guarantee it.

    >The molecular weight of the antibody itsself will be ˜150 kDa plus anadditional 240 kDa from the PE fluorophore. The molecular weight detected by the antibody will depend on what protein is His-tagged. The 6X His tag will add < 1 kDa to the weight of the protein, so the band should appear at the same molecular weight as the untagged protein.
