The antibody has been tested at transmission electron microscopy in human adipocytes fixed with paraformaldehyde, trated with NH4Cl to block free aldehyde groups, dehydrated with graded concentrations of ethanol and embedded in acrylic(either LRWhite or Lowicryl K4M) resins. Embedding in Lowicryl K4M resin excluded thermic denaturation of the antigen since the whole processing occurs at low temperature.
The antibody was tested diluted (from 1:20 to 1:5) as well as undiluted, but the result was always the same: an evident signal on chromatin (Ch) in the nuclei (N), some signal in the cytoplasm, a negligible signal around lipid droplets (L).
