  • SK-N-BE cells labeling Piccolo using ab101654 (green) at 1/100 dilution in ICC/IF. Cells were fixed using 4% formaldehyde for 15 minutes at room temperature. Primary antibody was incubated for 1 hour at room temperature. Secondary antibody was a goat anti-mouse ATTO 488 at 1/100 dilution for 1 hour at room temperature. Counterstained with Phalloidin Texas Red F-actin stain. Nuclei were stained with DAPI (blue).

Yes, ab101654 is the only mous monoclonal antibody we have for Piccolo. It has not yet been tested with mouse samples and in IHC-P (paraffin-embedded PFA-fixed tissue sections). We cannot guarantee if and how well the antibody will work, but you can use our testing discount program to try the antibody.

Vielen Dank für Ihren Anruf. Wie versprochen hatte ich für den anti-Piccolo- Antikörper ab101654 nachgefragt, ob das Epitop kartiert worden ist. Daraufhin hat man mir die Immunogensequenz geschickt, die zur Generierung dieses Antikörpers verwendet wurde (siehe Anhang). Es handelt sich dabei um die 356 Aminosäuren lange Isoform 3 des humanen Piccoloproteins (Genbank Accession number BC001304.1, SwissProtID Q9Y6V0, http://www.uniprot.org/uniprot/Q9Y6V0#Q9Y6V0-3). Ein Alignment der verschiedenen Isoformen und Sequenzen zeigt, dass Isoform 3 den C-terminalen Aminosäuren (ab ca Position 4500) der Isoformen 2, 5 und 6 entspricht. Schließt dies Ihre geplante Mutation ein und würde sich der Antikörper damit für Ihre Vorhaben eignen? Sie erwähnten auch, dass Sie eventuell IP-Experimente durchführen möchten. Unseres Wissens nach wurde der Antikörper ab101654 bisher noch nicht in IP getestet.
