    Thank you for getting back to me. Thank you also for trying my suggestions. I am sorry that they did not lead to any improvements to the results that you have been obtaining. As mentioned previously unfortunately we cannot guarantee this antibody to work in mouse as it is yet to be tested. I can only suggest that you try applying this antibody to a positive control tissue such as rat brain. I am sorry that I cannot be of more assistance on this occasion.

    Thank you for your enquiry. I am sorry to hear that you have been having difficulties with this antibody. As my colleague has explained unfortunately this antibody is yet to be tested using mouse and human tissue. However, I would predict its reactivity given 100% sequence similarity. I would like to suggest that you apply the antibody using an overnight incubation at 4oC using a reduced dilution e.g. 1:100.     Thank you for the protocol details that you have provided and I'm sorry to hear that you're experiencing difficulty with ab4138. I noticed that you are using human and mouse samples. Ab4138 has been tested for cross-reactivity with rat PKC beta 2 and although it is predicted to cross-react with other mammals dues to sequence homology, it hasn't been tested yet and we can't guarantee results. I checked with the originator of this antibody and the antibody was tested in Western blotting using rat brain, heart, lung and spleen with the highest expression occurring in brain and spleen. I would therefore suggest using rat brain or spleen as a positive control. If this does not help, and you have purchased the antibody in the past 90 days, I would be happy to offer you a free of charge replacement or refund.

    Thank you for your enquiry and for your detailed protocol. This antibody recognizes Rat PKC beta 2 and the predicted molecular weight is kDa. We would like to emphasize the fact that this antibody has only been tested on rat samples. Since you are using human, mouse and hamster samples these cell lines may show different pattern on the Western blot. You may also need to consider the fact that ab4138 is a polyclonal antibody, therefore it recognizes different epitopes. We would strongly suggest running rat samples as positive control along with the cultured cell lines. You may also need to test the secondary antibody to see if there are any non-specific bands due to the secondary antibody. Alternatively, you can decrease the concentration of the primary antibody and apply more stringent washes. Should you still have any problem, please do not hesitate to contact us again.
