    This is a calcium-independent, phospholipid-dependent enzyme specific for serine and threonine. PKC is activated by diacylglycerol which in turn phosphorylates a range of cellular proteins. PKC also serves as the receptor for phorbol esters, a class of tumour promoters.

Thank you for your enquiry. I am sorry but we do not recommend a positive control lysate for this antibody. However, following some literature searching I have found a publication where a purified form of the protein was used to demonstrate expression in endocardial and epicardial tissues in rat. Thank you for your e-mail regarding the antibody ab11724. I appreciate you taking the time to express your disappointment with this product, your opinion is very valuable to us and we take your comments very seriously. Thank you for the images which already give an insight into your problem, I can see that the first membrane shows a band just under 100kda which is likely to be PKC epsilon in the cytoplasmic fraction only (and is not seen in the membrane fraction) )and the predicted MW is 88kda) but also 2-3 other non specific bands. The second and third blots are similar in patterns but differ in intensity, which I think is a reflection of the secondary antibodies used. Non specific bands could be due to a number of reasons. As you see a nice single band with the phospho PKC epsilon antibody I suspect the problem is not due to the degradation of the samples. Could you please tell me how you block the membrane and what antibody dilution buffer you have used? This may give me an indication as to what the problem might be. Could you also tell me what cells were used and how the antibody was stored? Finally, if you could provide your order details I would like to see if there was a delay in the shipping of the tube, which would indicate a damaged vial. I look forward to hearing from you to resolve this matter.
