ELISA Use at an assay dependent concentration.
IHC-P 1/100. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.
WB 1/1000. Predicted molecular weight: 71 kDa. 1/1000 when using colorimetric substrates such as BCIP/NBT, and 1/5000 for chemiluminescent substrates. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. Dilution optimised using Chromogenic detection.


  • Function

    The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin.
  • Involvement in disease

    Defects in KLKB1 are the cause of prekallikrein deficiency (PKK deficiency) [MIM:612423]; also known as Fletcher factor deficiency. This disorder is a blood coagulation defect.

  • ab44392 staining Kallikrein 1B in human temporal neocortical tissue sections IHC-P . Tissue samples were fixed with formaldehyde and blocked with 10% serum for 20 minutes at 23°C: antigen retrieval was performed by heat mediation in citrate buffer.. Samples were incubated with primary antibody (1/100) for 18 hours at 23°C . A biotin-conjugated horse anti Rabbit IgG  polyclonal (1/50) was used as secondary antibody.

This is frustrating for us too, finding incorrect information on our datasheets. We try to minimize it but there is clearly work to be done.

I did not realize you had already purchased the antibody. If you would still like to try it with rat samples and it fails, I will offer a refund or credit or replacement with another antibody. Otherwise, I can send you instructions on how to return it for a full refund.

Of the other KLK1B antibodies, ab43084 and ab44203 have similar differences with the rat protein (8/35 and 10/35 residue mismatches respectively). It is unlikely these have been tested with rat tissues as they are from the same laboratory that provided ab44392. I am confirming with that lab.

The other two candidates are ab116517 and ab75228 but the latter appears to be detecting the heavy chain (as does ab1006). I will have the originating laboratory align the ab116517 immunogen with the rat KLK1B sequence, and will confirm if it is on the light chain.

Antibody ab1006 reactivity with rat was not confirmed in-house but was added to the datasheet based on the second reference listed on the datasheet, which shows a blot in figure 5A.

My apologies: the laboratory that supplies this to us confirmed that they have not tested this for reactivity to rat protein, for have we or, to our knowledge, any of our customers. It is not clear why our datasheet lists rat in the "Reacts With" field but I am assuming there was a miscommunication.

I am going to remove rat reactivity from the datasheet, so it will not be covered by our guarantee. If you are still interested in trying it, we do offer a free antibody in return for a review of an antibody tested in previously untested species or application.

In brief, you would buy ab44392 at full price, test it against your rat samples in one of the listed tested applications (ELISA, IHC-P, WB), and send us a review via our "Abreview" program.
