Target

  • Function

    The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin.
  • Involvement in disease

    Defects in KLKB1 are the cause of prekallikrein deficiency (PKK deficiency) [MIM:612423]; also known as Fletcher factor deficiency. This disorder is a blood coagulation defect.

    Hello,

    Can you tell me what is the actual quantity (e.g. in micrograms) of antibody that is included in the rabbit monoclonal anti-plasmakallikrein1B (ab124938)?

    The size says only 100 ul, and while it gives recommended dilutions for some purposes, I want to use it for another purpose-- i.e. to covalently conjugate it to magnetic beads for a bead-based IP, and for this process, I need to know the absolute quantity of antibody. Thanks,

    Answer

    Thank you for contacting us with your question.

    Ab124938 is provided as unpurified tissue culture supernatant, and since there other proteins present in this solution, it isn't possible to measure the concentration of the specific antibody. So unfortunately we do not know the mass of antibody in the 100 uL provided. Each new lot is tested and adjusted so that the optimal results are seen using the dilutions listed on the datasheet.

    It is possibleto purify the IgG molecules out of the supernatant using a protein A or G purification and then measuring the concentration, however the yield will be very low with only 100 uL (we recommend 10-50 mL for our supernatant purification kits like ab109207). So, if the mass or concentration is essential information for your assay, I would recommend using a purified antibody.

    We have several other plasma kallikrein 1B antibodies such as ab1006 and ab44392, and though these haven't been validated in IP they may work in this application. We do have a testing discount program if you'd be interested in trying on of these other antibodies in your application.
