    Ab247772 is the carrier-free version of ab109124. This format is designed for use in antibody labeling, including fluorochromes, metal isotopes, oligonucleotides, enzymes.

     

    Our carrier-free formats are supplied in a buffer free of BSA, sodium azide and glycerol for higher conjugation efficiency.

Application Abreviews Notes
ICC Use at an assay dependent concentration.
IHC-P Use at an assay dependent concentration. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.

For antigen retrieval, heat up to 98 degree C, below boiling, and then let cool for 10-20 minutes.

 

WB Use at an assay dependent concentration. Predicted molecular weight: 70 kDa.

    • Function

      Actin-binding protein. Plays a role in the activation of T-cells in response to costimulation through TCR/CD3 and CD2 or CD28. Modulates the cell surface expression of IL2RA/CD25 and CD69.
    • Tissue specificity

      Detected in intestinal microvilli, hair cell stereocilia, and fibroblast filopodia, in spleen and other lymph node-containing organs. Expressed in peripheral blood T lymphocytes, neutrophils, monocytes, B lymphocytes, and myeloid cells.
    • Sequence similarities

      Contains 2 actin-binding domains.
      Contains 4 CH (calponin-homology) domains.
      Contains 2 EF-hand domains.
    • Post-translational
      modifications

      Phosphorylated on a serine residue in response to costimulation through TCR/CD3 and CD2 or CD28. Serine phosphorylation promotes association with the actin cytoskeleton and targeting to peripheral cell projections.
      The N-terminus is blocked.
    • Cellular localization

      Cytoplasm > cytoskeleton. Cell junction. Cell projection. Cell projection > ruffle membrane. Relocalizes to the immunological synapse between peripheral blood T lymphocytes and antibody-presenting cells in response to costimulation through TCR/CD3 and CD2 or CD28. Associated with the actin cytoskeleton at membrane ruffles (By similarity). Relocalizes to actin-rich cell projections upon serine phosphorylation.
