Our Abpromise guarantee covers the use of ab201241 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
This product gave a positive signal in K562 cells fixed with 4% formaldehyde (10 min) and 80% methanol (5 min).
FunctionThe production of the second messenger molecules diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3) is mediated by activated phosphatidylinositol-specific phospholipase C enzymes. It is a crucial enzyme in transmembrane signaling.
Sequence similaritiesContains 1 C2 domain.
Contains 1 PH domain.
Contains 1 PI-PLC X-box domain.
Contains 1 PI-PLC Y-box domain.
Contains 2 SH2 domains.
Contains 1 SH3 domain.
modificationsPhosphorylated on tyrosine residues; upon ligand-induced activation of a variety of growth factor receptors and immune system receptors. Increases phospholipase activity.

ab201241 staining PLCG2 in K562 cells. The cells were fixed with 80%methanol, permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab201241 at 1/100 dilution (shown in green) and ab195889, Mouse monoclonal [DM1A] to alpha Tubulin - Microtubule Marker (Alexa Fluor® 594), at 1/250 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
