Inquiry: Hi, I am looking for the antibody against PLEKHM2 (ab91581). According to WB results on the datasheet, it detected a band at size around 169 kDa. However, MW of PLEKHM2 will be only about 110 kDa according to its human amino acid sequence. I am wondering how you can resolve or explain this difference. Cheers,

Thank you for your inquiry.
The band size of proteins on a WB is dependent o0n many factors like percentage of the gel and pH of the puffers, etc.
Especially big protein are difficult to measure precisely on gels since the marker bands are also affected by these factors and might be affected differently form the analyzed protein.
PLEKHM2 is also modified on some amino acid residues. This will add to the molecular weight.
The WB images shows that the band disappears when the antibody is pre-incubated with the blocking peptide. This indicates that the band is specific.
I hope this information is helpful and wish you good luck with your research.
