  • Function
    May be involved in cell migration and/or actin cytoskeleton organization. When expressed in keratinocytes, induces changes in cell morphology with transfected cells showing an elongated shape, numerous membrane protrusions, major reorganization of the actin cytoskeleton, increased motility and decreased cell adhesion. Required for normal lung cell proliferation and alveolus formation at birth. Induces platelet aggregation. Does not have any effect on folic acid or amino acid transport. Does not function as a water channel or as a regulator of aquaporin-type water channels.

    Highly expressed in placenta, lung, skeletal muscle and brain. Weakly expressed in brain, kidney and liver. In placenta, expressed on the apical plasma membrane of endothelium. In lung, expressed in alveolar epithelium. Up-regulated in colorectal tumors and expressed in 25% of early oral squamous cell carcinomas.

  • ab11936 staining Podoplanin / gp36 in murine skin tissue sections by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).
    Tissue was fixed in paraformaldehyde and an enzymatic antigen retrieval step was performed using Protein K. Samples were then blocked with 5% serum for 30 minutes at room temperature and then incubated with ab11936 at a 1/400 dilution for 16 hours at 4°C. The secondary used was a biotin conjugated goat anti-hamster IgG used at a 1/500 dilution.

  • ab11936 staining Podoplanin / gp36 in mouse lung tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with formaldehyde. Samples were incubated with primary antibody (1/80 in PBS + 1% BSA) for 16 hours at 4°C. A Biotin-conjugated goat anti-hamster IgG polyclonal (1/500) was used as the secondary antibody.

    Podoplanin is revealed in green (Alexa Fluor® 488) and nucleus are counterstain in blue (DAPI). Streptavidine-Alexa Fluor® 488 was used to increase signal.

  • Immunohistochemistry (PFA perfusion fixed frozen sections) analysis of mouse liver tissue sections labeling Podoplanin / gp36 with ab11936 at 1/250 dilution. Cells were fixed with formaldehyde, followed by blocking with PBS + 0.1% Triton X-100, 1% BSA, 10% goat serum for 1 hour at 25°C. A polyclonal goat anti-hamster Alexa Fluor 488 secondary antibody was used at 1/500 dilution.

Here is a link to a Histonet post which you may have already seen. The advice is generally to use clean, positively-charged slides (Superfrost Plus from Fisher is mentioned often, here and in other posts), and very specific instructions to warm the slides before de-paraffinization to remove all traces of water. (It improves adhesion later on in the procedure).

I was not able to find any articles in the Journal of Immunohistochemisty or Journal of Histotechnology that had other advice. You may want to try experimenting with enzymatic retrieval, such as trypsin or proteinase K, though we do not have data for this approach with this antibody.

I can confirm that the clone number of ab11936 is RTD4E10 which can be found in the brackets of the target name as well as on the on-line product datasheet under the clone number.

ab11936:Anti-Podoplanin / gp36 antibody - [RTD4E10]

Both antibodies recognize murine Podoplanin, however ab92319 has only been characterized in IHC-Fr.

On the other hand, ab11936 has been tested in different types of immunohistochemistry ie.e IHC-FoFr, IHC-Fr, IHC-P. If you have used ab11936 successfully in the past and are satisfied with it, I would recommend performing the staining experiments with this product, though the final decision is yours.

As discussed on the telephone, far as we are aware, ab10288 Podoplanin / gp36 antibody [18H5] has never been tested with samples from mouse. All tested species cross-reactivity information is stated on our datasheets, and these are updated as soon as any new information is brought to our attention.

I have reviewed the alignment of the immunogen for this antibody with the mouse protein, and regrettably this is low at 39%. Therefore, we would not expect this antibody to detect in mouse samples. I'm sorry this is not better news for you.

I can recommend that ab11936 Anti-Podoplanin / gp36 antibodywould be amore suitable option as it is already tested in mouse. You will need to ensure it is tested in the application you are using so it is covered by the guarantee for your experiments.
