Our Abpromise guarantee covers the use of ab168371 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|IP||1/10 - 1/100.|
|ICC/IF||1/50 - 1/100.|
|Flow Cyt||1/100 - 1/500.
ab172730 - Rabbit monoclonal IgG, is suitable for use as an isotype control with this antibody.
|WB||1/10000 - 1/50000. Detects a band of approximately 36 kDa (predicted molecular weight: 36 kDa).|
FunctionPP2A can modulate the activity of phosphorylase B kinase casein kinase 2, mitogen-stimulated S6 kinase, and MAP-2 kinase.
Sequence similaritiesBelongs to the PPP phosphatase family. PP-1 subfamily.
modificationsReversibly methyl esterified on Leu-309. Carboxyl methylation may play a role in holoenzyme assembly. It varies during the cell cycle. Demethylated by PME1 (in vitro).
Phosphorylation of either threonine (by autophosphorylation-activated protein kinase) or tyrosine results in inactivation of the phosphatase. Auto-dephosphorylation has been suggested as a mechanism for reactivation.
Cellular localizationCytoplasm. Nucleus. Chromosome > centromere. Cytoplasm > cytoskeleton > spindle pole. In prometaphase cells, but not in anaphase cells, localizes at centromeres. During mitosis, also found at spindle poles.

ab168371 was shown to specifically react with PPP2CB in wild-type HAP1 cells. No band was observed when PPP2CB knockout samples were examined. Wild-type and PPP2CB knockout samples were subjected to SDS-PAGE. Ab168371 and ab18058 (Mouse anti-Vinculin loading control) were incubated overnight at 4°C at 1/10,000 dilution and 1/10,000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/10,000 dilution for 1 hour at room temperature before imaging.
Detection of PPP2CB by Western Blot of Immunprecipitate.
