      PP2A can modulate the activity of phosphorylase B kinase casein kinase 2, mitogen-stimulated S6 kinase, and MAP-2 kinase.

      Reversibly methyl esterified on Leu-309. Carboxyl methylation may play a role in holoenzyme assembly. It varies during the cell cycle. Demethylated by PME1 (in vitro).
      Phosphorylation of either threonine (by autophosphorylation-activated protein kinase) or tyrosine results in inactivation of the phosphatase. Auto-dephosphorylation has been suggested as a mechanism for reactivation.

      Cytoplasm. Nucleus. Chromosome > centromere. Cytoplasm > cytoskeleton > spindle pole. In prometaphase cells, but not in anaphase cells, localizes at centromeres. During mitosis, also found at spindle poles.

      ab168371 was shown to specifically react with PPP2CB in wild-type HAP1 cells. No band was observed when PPP2CB knockout samples were examined. Wild-type and PPP2CB knockout samples were subjected to SDS-PAGE. Ab168371 and ab18058 (Mouse anti-Vinculin loading control) were incubated overnight at 4°C at 1/10,000 dilution and 1/10,000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/10,000 dilution for 1 hour at room temperature before imaging.

    • Detection of PPP2CB by Western Blot of Immunprecipitate.
      293T cell lysate immunoprecipitated using ab168371 at 1/10 dilution.
      For WB detection ab168371 at 1/10000 dilution was used.
      HRP-conjugated anti-rabbit IgG preferentially detecting the non-reduced form of rabbit IgG.

    • Flow Cytometric analysis of permeabilized NIH 3T3 cells labeling PPP2CB with ab168371 at 1/100  dilution (red) or a rabbit IgG (negative) (green).

    • Immunofluorescent analysis of 293T cells labeling PP2A beta with ab168371 at 1/50 dilution.

    • All lanes : Anti-PP2A beta antibody [EPR11786(B)] (ab168371) at 1/10000 dilution
