  • Function
    Protein phosphatase 1 (PP1) is essential for cell division, and participates in the regulation of glycogen metabolism, muscle contractility and protein synthesis. Involved in regulation of ionic conductances and long-term synaptic plasticity. May play an important role in dephosphorylating substrates such as the postsynaptic density-associated Ca(2+)/calmodulin dependent protein kinase II. Component of the PTW/PP1 phosphatase complex, which plays a role in the control of chromatin structure and cell cycle progression during the transition from mitosis into interphase.

    Cytoplasm. Nucleus. Nucleus > nucleoplasm. Nucleus > nucleolus. Primarily nuclear and largely excluded from the nucleolus. Highly mobile in cells and can be relocalized through interaction with targeting subunits. NOM1 plays a role in targeting this protein to the nucleolus. In the presence of PPP1R8 relocalizes from the nucleus to nuclear speckles.

We have received no complaints about this product, therefore I am concerned it is not working for you in IP, although it is fine in Western blot. Unfortunately, we currently do not have a different lot of this PPP1A antibody available. Therefore, I would like to make some suggestions to the protocol in an attempt to improve the results obtained in IP. I would also appreciate if you can confirm some details of the procedure. 1) Is 2 µg/ml the optimised antibody concentration? The amount of antibody used needs to be optimised depending on the abundance of the protein and the affinity of the antibody for the protein as well as the method used: The first approach is to mix antibody with protein sample and allowing it to interact with protein followed by addition of Protein A/G support. The second approach (which I understand you have used) is to bind antibody to the Protein A/G beads and then mix with the antigen.This method gives lesser yield than first one and may require more antibody. Please find attached our detailed protocol guide. 2) Could you please confirm which positive control you have used, in order to check the Protein A-agarose and other reagents? 3) Which elution buffer have you used and for how long have you incubated to elute the proteins from the beads? I hope optimisation of the IP conditions will improve the results. As the antibody was purchased just recently you are covered by our Abpromise guarantee. Therefore if the suggestions do not improve the results I would be happy to offer a refund or replacement vial of a primary antibody of your choice.
