    The PR65 subunit of protein phosphatase 2A serves as a scaffolding molecule to coordinate the assembly of the catalytic subunit and a variable regulatory B subunit. Required for proper chromosome segregation and for centromeric localization of SGOL1 in mitosis.

Here is the IHC testing protocol that was used.

The laboratory used Tris-EDTA buffer as antigen retrieval, but the suitable buffer may be dependent on different tissue. We recommend the customer try Tris-EDTA buffer and citrate buffer, and optimize the condition based on the testing result.

IHC protocol

1. Heat the paraffin-embedded tissue sections in 60℃ oven for 90mins.

2. Antigen retrieval /deparaffine: Incubate the sections in antigen retrieval buffer (EDTA-based, pH 8.0) in high pressure Cooker for 15mins.

3. PBS wash twice for 5 mins.

4. After the sections cools down, Incubate the sections in 3% H2O2 at RT for 30min.

5. PBS wash twice for 5 mins.

6. Incubate the sections in 1.5% goat serum (dilute goat serum by 1xPBS) at RT for 30

mins.

7. Incubate the sections in primary antibody at 4 degree C for overnight. (dilute primary antibody in 1.5% goat serum blocking buffer, (dilutions depends on the antibody)

8. PBS wash twice for 5 mins.

9. Incubate the sections in secondary antibody at 1:200 dilution (dilution depends on the antibody) at RT for 30min. (dilute the primary antibody in blocking buffer)

10. PBS wash twice for 5 mins.

11. Add ABC reagent for 30mins at RT

12. PBS wash twice for 5 mins.

13. Add DAB reagent until the signal appears (within 5 mins)

14. PBS wash twice for 5 mins.

15. Counter-stain the section in Hematoxylin at RT for 30 sec ˜ 3 mins.

16. Wash the section by running tap water for 5 mins.

17. Dehydrate sections:

a. Incubate the section with 95% EtOH at RT for 5 mins.

b. Incubate the section with 99.5% EtOH at RT for mins.

c. Incubate the section with Xylene for at RT for 5 mins.

18. Mount the sections with coverslip.
