    • Lane 1: Wild-type HAP1 cell lysate (20 µg)
      Lane 2: PPP2R1B knockout HAP1 cell lysate (20 µg)

      Lanes 1 - 4: Merged signal (red and green). Green - ab154815 observed at 67 kDa. Red - loading control, ab8245, observed at 37 kDa.

      ab154815 was shown to specifically react with PPP2R1B when PPP2R1B knockout samples were used. Wild-type and PPP2R1B knockout samples were subjected to SDS-PAGE. ab154815 and ab8245 (loading control to GAPDH) were diluted at 1/1000 and 1/10 000 respectively and incubated overnight at 4°C. Blots were developed with goat anti-rabbit IgG (H + L) and goat anti-mouse IgG (H + L) secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

    • Flow Cytometry analysis of Caco-2 (human colorectal adenocarcinoma ) cells labeling PPP2R1B with unpurified ab154815 at 1:40 dilution(10ug/ml) (red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. A Goat anti rabbit IgG (Alexa Fluor® 488)(ab150077)(1:2000 dilution) was used as the secondary antibody. Rabbit monoclonal IgG (Black)(ab172730) was used as the isotype control, Cell without incubation with primary antibody and secondary antibody (Blue) were used as the unlabeled control.

    Customer reviews and Q&As


    We have not tested 7288-1 or 7301-1 for cross reactivity with PP2AA alpha, however, we have only observed one band in our in-house testing with 7288-1 and we have detected only 2 isoforms of PP2AA beta with 7301-1. It is possible they could react, based on the fact that the immunogen sequences for 7288-1 and 7301-1 have 78% and 75% homology with PP2AA alpha (respectively), but typically homology needs to be at about 85% for there to be good cross-reactivity.
