Thank you for your enquiry. Our source for this antibody was able to kindly provide me with the following information. "Based on an amino acid sequence comparison, I did not find any homology between the stated immunogen sequence (Arg-Glu-Pro-Ser- Lys-Asn-Ala-Pro-His-Ser-Gln-Gly-Glu) and the sequence of gamma isoform of regulatory subunit B56, protein phosphatase 2A(isoform 4), as found on the PubMed Protein database. It would appear then that the antibody is binding elsewhere on the PP2A/B gamma molecule. However, my discussion with the manufacturer ended with his saying he had found the immunogen sequence within the B55 subunit, but can't be certain that that is where antibody binds due to the complexity of the molecule. And indeed, complex it is. I found this description on the Internet: "PP2A consists of a common heterodimeric core enzyme, composed of a 36 kDa catalytic subunit (subunit C) and a 65 kDa constant regulatory subunit (PR65 or subunit A), that associates with a variety of regulatory subunits. Proteins that associate with the core dimer include three families of regulatory subunits B (the R2/B/PR55/B55, R3/B'/PR72/PR130/PR59 and R5/B'/B56 families), the 48 kDa variable regulatory subunit, viral proteins, and cell signaling molecules (By similarity)." The final answer is that I don't think anyone knows for sure where this antibody binds, with some indication that the B55 regulatory subunit might be it."
