  • Function

    Catalytic subunit of protein phospatase 6 (PP6). PP6 is a component of a signaling pathway regulating cell cycle progression in response to IL2 receptor stimulation. N-terminal domain restricts G1 to S phase progression in cancer cells, in part through control of cyclin D1. Downregulates MAP3K7 kinase activation of the IL1 signaling pathway by dephosphorylation of MAP3K7.
  • Tissue specificity

    Ubiquitously expressed in all tissues tested with highest expression levels in testis, heart, kidney, brain, stomach, liver and skeletal muscle and lowest in placenta, lung colon and spleen.
  • Sequence similarities

    Belongs to the PPP phosphatase family. PP-6 (PP-V) subfamily.

    Hi:

    To answer your questions:

    1. The buffer is what I use, the lysis buffer is that with 1% triton (added before the lysis).

    2. Yes, it is a very good antibody

    3. I probed the sample for another band (a phospho antibody and am probing for loading control now, but the last blot which also has not worked, has been probed for a protein called PAK1 and actin)

    4. The Odyssey Licor appratus was used at other power settings as well as highest power.

    I will try to help as best I can to resolve these problems. The information provided has been very helpful in understanding the experiment you have been performing. I do have a few follow up questions to ask just to be able to ascertain what may be contributing to this lack of signal from the samples.

    1. How were the lysates prepared? The buffer composition you shared, was this used as the lysis buffer as well as the SDS-page loading buffer?

    2. Has the secondary antibody (goat anti-rabbit Alexa Fluor 680) been used previously with a different primary antibody? How were these results?

    3. Was any loading control used with the samples? For example actin or GAPDH probed? What was the results of this?

    4. What exposure was used on the membrane?

    I have a question about an antibody cross-species specificity. I found antibodies against human PP6 in your catalog: ab60249 and ab70634. I am wondering whether these antibodies are able to recognize Drosophila (fruit fly) PP6 protein. Do you have any information about that? Although the PP6 protein is highly conserved between human and fly species, I am still not quite sure whether your antibodies will work or not in fly system. Therefore, I want to get/purchase a small size sample to test in fly. Is it possible? Thank you in advance for your help.
