    Specific for eosinophil major basic protein (14kDa) irrespective of the stages of eosinophil activation and can be regarded as a “pan-eosinophil” marker. Recognizes secreted pro-form of MBP (Pregnancy associated MBP, 23.8kDa) and mature form found in the matrix of the eosinophil large specific granule (13.8kDa). The clone number has been updated from (1.B.787) to (BMK13) both clone numbers name the same antibody clone.

  • Application notes
    ELISA: Use at an assay dependent dilution.
    IHC-P: Use at an assay dependent dilution. Perform enzymatic antigen retrieval before commencing with IHC staining protocol (0.1% trypsin).
    IHC-Fr: Use at an assay dependent dilution. Fix with methanol:acetone.
    WB: Use at an assay dependent dilution. Predicted molecular weight: 26.6 kDa.

    Not tested in other applications.
    Optimal dilutions/concentrations should be determined by the end user.
  • Target

    • Function

      Cytotoxin and helminthotoxin. Also induces non-cytolytic histamine release from human basophils. Involved in antiparasitic defense mechanisms and immune hypersensitivity reactions. The proform acts as a proteinase inhibitor, reducing the activity of PAPPA.
    • Tissue specificity

      High levels of the proform in placenta and pregnancy serum; in placenta, localized to X cells of septa and anchoring villi. Lower levels in a variety of other tissues including kidney, myometrium, endometrium, ovaries, breast, prostate, bone marrow and colon.
    • Sequence similarities

      Contains 1 C-type lectin domain.
    • Developmental stage

      Levels of the proform increase in serum and placenta during pregnancy.
    • Post-translational
      modifications

      Nitrated.
    • Cellular localization

      Cytoplasmic vesicle > secretory vesicle. The proform is secreted. The mature protein is found in the matrix of the eosinophil's large specific granule and Secreted. The proform is secreted.

      I have received confirmation from the laboratory that tested ab14462 that your protocol is very good and should work with ab14462. The laboratory shared also the following feedback which may interest you "Your customer's secondary antibody is a Sheep x mouse IgG HRP. Ab14462 is a mouse IgG1. I have had diverse experiences with this sort of distinction. I have had customers state that an anti-IgG will recognize all IgG's of the same species; and yet there have been times when I suggested matching the secondary precisely to the target isotype and suddenly the protocol was working." As the antibody did not work for you I would be happy to offer you a replacement vial or credit note/refund.       I have been informed by the source of ab14462 that it was not tested in house in ELISA and that unfortunately we therefore do not know the recommended dilution. I was recommended to start at a concentration of 2-5ug/ml (for Direct or Indirect ELISA), incubating overnight at 4C. I'm sorry I cannot provide further help. I will look into your complaint as soon as possible and appreciate your patience,
