  • Function

    Arginine methyltransferase that methylates (mono and asymmetric dimethylation) the guanidino nitrogens of arginyl residues present in proteins such as ESR1, histone H2, H3 and H4, PIAS1, HNRNPA1, HNRNPD, NFATC2IP, SUPT5H, TAF15 and EWS. Constitutes the main enzyme that mediates monomethylation and asymmetric dimethylation of histone H4 'Arg-4' (H4R3me1 and H4R3me2a, respectively), a specific tag for epigenetic transcriptional activation. Together with dimethylated PIAS1, represses STAT1 transcriptional activity, in the late phase of interferon gamma (IFN-gamma) signaling. May be involved in the regulation of TAF15 transcriptional activity, act as an activator of estrogen receptor (ER)-mediated transactivation, play a key role in neurite outgrowth and act as a negative regulator of megakaryocytic differentiation, by modulating p38 MAPK pathway.

  • Western blot analysis (whole HL60 cell lysate): PRMT1 (Arrow) was detected using ab3768 (Lane B) but not pre-immune serum (lane A). Secondary HRP-anti-rabbit was used for signal visualization with chemiluminescence.
    Sonicated Chromatin prepared from untreated or 17beta-estradiol (E2) treated MCF7 cells was subjected to the ChIP procedure with ab4441 to Histone H3 (acetyl K9) and the immunoprecipitated chromatin was analysed in the proximal region of the estrogen-responsive pS2 promoter (as shown above) and quantified by real-time PCR (values are % of inputs). The primers are designed to follow the nucleosome E (including the Estrogen Responsive Element ERE).
    10 µl of ab3768 and 2x10

    Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human breast carcinoma tissue labeling PRMT1 - ChIP Grade with ab3768. Tissue was fixed with formaldehyde and blocked with 3% BSA for 0.5 hour at 37°C; heat mediated antigen retrieval was performed with a citrate buffer (pH6). Samples were incubated with primary antibody (1/25) for 1 hours at 37°C. A Peroxidase-conjugated Goat anti-rabbit polyclonal (ready to use) was used as the secondary antibody.

I am very sorry for the delay in replying to you and for the problems you are having with those antibodies in ChIP. We have not tested in house those antibodies in ChIP and therefore it is very difficult to find what the problem might be. Ab5408 (polII) has been tested in ChIP by a customer who sent us a review saying it worked in ChIp for him. You can e-mail the reviewer by clicking on his name after finding his review on the reviews tab. He hopefully will be able to help you. Ab651 had been tested in ChIP by a collaborator but having discussed with my colleagues your problem we have contacted another collaborator who expressed his concerns as to the amount of antibody needed to do the IP and hence we have decided today to remove the ChIP grade. I am very sorry for the inconvenience and can assure you we can offer you a refund if you wish. Ab3758 was also tested in ChIP by the same collaborator and he told us "I think ab3768 is working in ChIP, the results are quite nice and correspond to what you expect. Indeed you expect and early recruitment of PRMT1 on the ps2 gene as described by Frank Gannon et al in their Cell paper." These results are found on the datasheet. He recommends 10ul of antibody for and 2x10E6 MCF7 cells treated with 17beta-estradiol. Unfortunately we are unable to find out more protocol information for those antibodies but I can certainly offer you a replacement vial of each, or a credit note or refund. Please let me know which you would prefer and I will arrange accordingly,
