      Regarding the ANT kit ab109713, I understand you would want to use at least 5uL of beads.
      Just to clarify, is that the amount you would recommend if we use the amount of mitchondria specified in the protocol ? Or more ?
      Also, exactly how many uL of beads are in the kit ? It looks like it is 25uL in the 250ug kit, which would mean that you can assay 5 samples with that kit (provided you use 5uL of beads). Is that correct ?

      Thank you for your reply.

      The kit contains 25 uL (settled volume) of beads conjugated to 250 ug of antibody. This is enough to run 5 tests ifyouuse5 uL beads for each test. The 5 uL recommended minimum is for any sample, including mitochondria. We recommend to use 5 uLof beads as the minimum amount, and you can try to use less amount of beads, but according to our experience, it is difficult to handle and easy to lose the beads during the procedure.But please keep in mind thatthe volume we talk about is the solid beads volume. It is not the volume of the slurry.

      Regarding kits ab 110168 and 109713 Hello, I am looking to purchase the Mitochondrial Isolation kit for tissues and the ANT immunocapture kit and I have a few questions. We would like to isolate mitochondria from frozen mouse skeletal muscle for western blotting as well as performing an ANT immunocapture with your kit ab109713. 1. Can you tell me if the mitochondria obtained from kit 110168 are ok to use with kit 109713? 2. How many samples can be assayed with the 250ug kit for the ANT immunocapture ? 3. How much starting muscle sample would you recommend we use to get enough mitochondria for the subsequent ANT immunocapture ? Thank you

      Yes, the Mitochondria Isolation Kit for Tissue (ab110168) is an ideal kit to use to isolate mitochondria for use with the ANT Immunocapture Kit (ab109713).

      For the ANT protocol, "The smallest working amount of beads is around 5 μl of solid beads in a small microtube (e.g. a 500 μl tube). It is not practical to use a volume smaller than this for immunocapture procedure, therefore it is recommended that when wishing to use less beads researchers dilute the concentrated Abcam’s beads with unloaded agarose beads, giving a workable volume of beads. The yield from such diluted beads will of course be less. Plain beads for dilution are available from Abcam."

      For muscle mitochondria, the minimal starting about is 200 ug and the recommended starting amount is 2-5 mg.

      ab109864 anti-ANT beads uses the anti-ANT monoclonal antibody ab110322. This antibody is specific to Mouse, Rat, Cow, Human, Caenorhabditis elegans ANT.

      The antibody was raised against full length recombinant human ANT, but the exact binding site is unknown.

      I blasted human and mouse and there is a 95% identity - so that is excellent similarity.
