    This rapid multiplexing microplate kit is used to determine both the activity and quantity of cytochrome c oxidase in a human sample with greater speed and simplicity. The ratio of the two is a measure of specific activity. The COX enzyme is immunocaptured within the wells of the microplate and the activity is determined by following the oxidation of reduced Cytochrome c colorimetrically by the absorbance change at 550 nm. Subsequently, in these same wells the quantity of the enzyme can then be determined by adding a COX specific detector antibody and the appropriate alkaline phophatase conjugated antibody; this phosphatase changes a substrate from colorless to yellow at 405 nm. This reaction takes place in a time dependent manner proportional to the amount of protein captured in the wells. Included in this kit for performance of the multiplexing assay are buffers, detergent, substrates, and 96-well microplate with monoclonal antibody pre-bound to the wells of the plate, allowing for a stream-lined assay.

Can the detergent for ab109910 be used in place of the extraction buffer for ab124535 and vice versa? Answer

Yes, we use 10% detergent in ab109910 which is available as ab109857 so the final is 1%. This detergent is pretty mild and yields native functional enzymes.

Also, a final of 1.5% could be used (i.e. extraction buffer) without significantly changing the results or activity yield.

“I would like to assay complex 4 activity in primary neurons after different treatments. On the first time I have used that kit I got some samples that show a first slope (increasing wthin the first minute) then a second slope decreasing fast and a third slope that is deceasing and maintained until the end.

Which slope should I take into account? the only slope that appears in all the samples is the last. Why do I get the first slope that increases?

Answer

The slope to measure is the fast decreasing slope. The cytochrome c oxidase activity assay measures the capacity of the immunocapture enzyme for oxidizing reduced cytochrome c. Reduced cytochrome c is measured at 550nm. A reduction in the reading means that the reduced cytochrome c is becoming oxidized.

Answer

We have not tested the enzyme activity portion of the assay (ab109910) after overnight incubation at 4C. My suggestion is to not do it and stick to the conditions set out in the protocol = 3 hours incubation of sample at RT – then perform the activity assay portion (this should be done on the same day the sample was extracted) – then leave overnight at 4C (with the activity buffer in the wells) and do the Sandwich ELISA portion of the assay the next day. With this format we know the assay performs optimally.

First, i'm doing cell culture with C2C12 cell line (mouse), i want to measure cytochrome C activity; i use 4 X10 5 cells /well in 12 well- PLATES, than i extract my proteins with RIPA buffer, my questions are :

1. Is there any kit to measure cytochrome C activity special mouse not human §

2. If yes , can i use this kit with my experimental conditions written above ( i mean with C2C12 cell line , the number of cell 4 X 10 power 5 cells )

3. This kit ; i must use it when i collect my cells with PBS ( cells will be in suspension ) or after i will extract my proteins using RIPA buffer ?

Note : i hade already recommanded mitochondria extraction kit from your compagny but i can't use it to extract mitochondria because my number of cells is so low and i can't increase it, so can i use a kit to measure cytochrome C activity with these conditions ;

2. Answer 2. Solution 1 does not contain succinate; instead the cytochrome c has been reduced with DTT and then purified.
