    Magnesium Assay Kit (ab102506) is a simple sensitive assay for quantitating magnesium in a variety of biological samples. The kit takes advantage of the specific requirement of glycerol kinase for Mg2+. An enzyme linked reaction leads to formation of an intensely colored (λmax = 450nm) product whose formation is proportional to Mg2+ concentration. The linear range of the assay is 2-15 nmoles with detection sensitivity ~40 µM.

    This product is manufactured by BioVision, an Abcam company and was previously called K385 Magnesium Colorimetric Assay Kit. K385-100 is the same size as the 100 test size of ab102506.

    Magnesium is the 11th most abundant element by mass in the human body. Mg2+ is essential to all living cells where it plays an important role in facilitating the processing of biological polyphosphates like ATP, DNA, RNA and enzyme functions. Mg2+ is the metallic ion at the center of chlorophyll, and a common additive to fertilizers. Mg2+ compounds are used as laxatives, antacids, and used to stabilize abnormal nerve excitation and blood vessel spasm i.e., eclampsia.

Question

I'm interested in the Calcium Colorimetric Assay.

I have a few technical questions about the assay and the materials.

According to the website the linear range of detection for this kit is between 0.1-25 mM? What about higher that?

Will other ions interfere with the measurements? Meaning presence of other divalent or monovalent ions?

In my solution there are also presence of MOPS ions and NaCl.  

What is the binding molecule that binds to the Ca ions? In Sigma it is the o-cresolphthalein molecule. Is the binding of your molecule overcome other interaction with other ions? Meaning that the binding is specific and the presence of other ions do not interfere with the measurement.

I'm also looking to measure the concertation of Mg and Zn. I found Magnesium Assay Kit, does it works with different binding molecule? Do they interfere with one another. Is it specific as well? What is the binding molecule in the Mg kit?

Is it possible to buy the binding molecule and not the entire kit?

1. According to the website the linear range of detection for this kit is between 0.1-25 mM? What about higher that?

It is possible to detect higher amount as well- however, we have not tested it and it may be above the accurate range of the assay.

2. Will other ions interfere with the measurements? Meaning presence of other divalent or monovalent ions?

ab102506 Magnesium Detection Kit did not give interference from other ions tested- Zn, Mn, Ni, Fe, Cu, Co, Ca

ab102505 Calcium Detection Kit (Colorimetric) not known to show interference from Fe+2 or Mg

3. In my solution there are also presence of MOPS ions and NaCl.  

In testing, for both kits ab102505 and ab102506 we didn’t see an issue with NaCl , however, MOPS was not tested.

4. What is the binding molecule that binds to the Ca ions? In Sigma it is the o-cresolphthalein molecule. Is the binding of your molecule overcome other interaction with other ions? Meaning that the binding is specific and the presence of other ions do not interfere with the measurement.

For the calcium assay ab102505 we have a proprietary buffer formulation that prevents interference from Fe and Mg.

6. What is the binding molecule in the Mg kit?

There are no binding molecules in the magnesium assay kit ab102506  - the assay is based on a reaction where Mg serves as a co-factor.


7. Is it possible to buy the binding molecule and not the entire kit?


Unfortunately, I am sorry this is not possible in this case.

Question

The enzyme mixture was reconstituted in 550 uL of assay buffer. It was stored at -20 C.

The standards were prepared by mixing 10 uL of magnesium standard with 990 uL of water. The remaining standards were diluted down from that stock solution.

Question

The kit was reconstituted 17-FEB-2012. I believe it was received a few days before that. The curve was repeated the following day with similar results. Once the enzyme mixture was added to the samples, all of the standards turned color at the same relative intensity. The kit/reagents have been stored at -20 C. Thanks for your help.

Thank you for your help. Here is the excel file for the data we generated. This was a standard curve. 1.1 mL of water was used to reconstitute the developer and 550 uL was used to reconstitute the enzyme. The rest of the protocol was followed. We repeated the curve on a second day and got similar results. Let me know if you have any ideas, please. Thanks.

Based on the data you show it looks like the issue is thatno reaction mix or old/expired mix was used as there is not even the hint of a difference between 0 and the rest of the standards.


How old is this kit? When was it first reconstituted? Storage temp after? Have you tried to repeat the assay fresh (not the replicates shown)?


Also, can you please provide a lot number and order number for this kit?
