    Thank you for contacting us. 
    This kit size refers to the amount of antibody.  For example, the 250 ug kit contains 250 ug of antibody conjugated to 25 uL of beads (settled volume) suspended in a total volume of 400 uL. Thus the concentration of the antibody is 10 ug per uL of beads, or 0.625 ug per uL of whole suspension.  The 250 ug, 500 ug, and 750 ug sizes can capture ˜13 ug, ˜26 ug, or ˜39 ug respectively of PDH complex from heart mitochondria. 
    For detecting Pyruvate Dehydrogenase E1-alpha, I would recommend either ab110330 or ab110334.  I have included all three sizes of the kit and both antibodies on the quote I sent you for your reference.  If you could let me know your shipping address and the products you are interested in, I will be happy to update the quote for you.

    Inquiry: Hi, I am interested in trying this product with rat samples. Do you have any direct evidence for its efficacy against rat PDH, do you know if it was raised against a sequence that is identical to rat? The product information states that it reacts with human and cow, and I am concerned that there are no "predicted" interactions despite the high sequence identity of PDH subunits between human, mouse and rat, etc.

    I have had the opportunity to speak with the lab regarding this on your behalf. They recommend using ab110333 (MSP06) with rat samples. The antibody targets on E2/E3 binding protein but pulls down the whole PDH complex using IP reagents and protocol. The product page for ab110333 is located at:

    Hello again,
    I had contacted you with a question concerning your PDH-capture antibody (ab109802 or MSP01) about half a year ago, and got a very good answer, so I was wondering if I could ask one more thing. I realize that it most likely is not possible to disclose the immunogen used in generation of this antibody but I was wondering if it's been verified that the antibody captures only the pyruvate dehydrogenase complex or whether there's a possibility of cross-reactivity with the branched-chain-alpha-ketoacid dehydrogenase, since they are very similar.

    Thank you for contacting us with your question, and for your patience while I have been in touch with the lab about the specificity of this antibody.
    My contact at the lab has responded:
    "We did mass spectrometry and immunoblotting using this antibody and did not see any other branched chain dehydrogenases represented such as OGDH subunits.Enzymatically they are quite similar, but the primary sequences are not that similar. E.g. PDH E2 – OGDH E2 are only 29% identical."

    Thank you for contacting us and for your interest in MSP01 (ab109802). I forwarded your email to the lab scientists at MitoSciences, and I received the following response: We do not know if the kinase and phosphatase are present, but if so then they are at low levels because we did not pick them up by mass spectrometry. We do know the enzyme is active.  Addition of ATP inhibits the enzyme activity which might suggest the presence of PDH kinases, however we do not see a corresponding increase in phosphorylation of PDH E1 alpha subunit. My thoughts are that they may be there in small amounts. I hope this information is helpful, but please let me know if you have any further questions.

    We haven't tested this kit with mouse samples so we unfortunately cannot provide any details. The kit has been optimized with cow and human samples so these samples are fully guaranteed.
