    Pyruvate Assay Kit (Colorimetric/Fluorometric) ab65342 provides a simple, direct and automation-ready procedure for measuring pyruvate concentration in various biological samples such as blood, cells, culture and fermentation media, etc.


    In the pyruvate assay protocol, pyruvate is oxidized by pyruvate oxidase via enzyme reactions to generate color (λ= 570 nm) and fluorescence (at Ex/Em = 535/587 nm). Since the color or fluorescence intensity is proportional to pyruvate content, the pyruvate concentration can be accurately measured.


    The kit detects 1-200 µM pyruvate.


    Pyruvate assay protocol summary:
    - add samples and standards to wells
    - add reaction mix and incubate for 30 min at room temp
    - analyze with microplate reader

    Pyruvate is a key intersection molecule in the netwrok of metabolic pathways. Pyruvate can be made from glucose through glycolysism converted back to carbohydrates via gluconeogenesis, or to fatty acids or energy via acetyl CoA. It can also be used to construct the amino acid alanine and be converted to ethanol. Glucose is converted to pyruvate which is then converted into acetyl CoA, the main input for the citric acid cycle (TCA). If insufficent oxygen is available, pyruvate breaks down anaerobically, creating lactate in animals and ethanol in plants and microorganisms.

Hi
I bought your Pyruvate assay kit to check the Pyruvate level in cells. I am missing instructions how to treat cells to be able to assay the pyruvate content as well as an idear about the cell number needed. Is your assay buffer an Lysis buffer or do I need to lyse the cells in another buffer first? Which reagent do interfere? Do you have examples using cell lines ?

Thank you for your enquiry regarding ab65342.

I apologize for my delayed response but was out of the office yesterday.

You can homogenize about 2 x 10^6 cells with 500 µl of the assay buffer, centrifuge it and take the supernatant for this assay. We would suggest using several dilutions/concentrations of your sample to ensure the readings are within the standard curve range. Due to the presence of LDH in serum, care must be taken during sample processing to prevent the conversion of pyruvate to lactate. Samples can be deproteinized by 10 Kd cutoff spin filter to remove proteins.

The assay kit ab65342 is capable of detecting pyruvate in human plasma. Please note, our products are for "Research Use Only" (RUO), and are not intended for diagnostic use. Also note that due to the presence of LDH in serum and plasma, care must be taken during sample processing to prevent the conversion of pyruvate to lactate. Samples can be deproteinized by 10 kDa cutoff spin filter (ab93349) to remove proteins.

Diluted the pyruvate standard to 1 nmol/microlit by adding 10 microlit of the standard to 990 microlit of pyruvate assay buffer and mixed well. Added 0, 2, 4, 6, 8, 10 microlit into a series of standard wells. Adjusted volume to 50 microlit/well with pyruvate assay buffer to generate 0, 2, 4, 6, 8, 10, 20 nmol/well of the pyruvate standard.



1st try:

Thank you for your inquiry.

I can confirm that this kithasbeen tested with urine, but not especially with urine samples that have been frozen.

I can suggest to run preliminary experiments to check what the difference in your samples will be when used frozen or fresh. This is especially recommended if you have many samples. A preliminary experiment will make sure that all will go well when testing abig number of samples simultaneously.

I assume that if you use samples that were all frozen first, there should be no problem. I suggest to freeze the sample quickly andto avoidfreeze/thaw cycles.

Pyruvate is a chemical compound and will be the same in human and mouse.

I am planning to resect tumours from mice and then measure multiple metabolites from their tissue lysates. With reference to the ATP assay kit (ab83355), pyruvate assay kit (ab65342), L-lactate assay kit (ab65331) and citrate assay kit (ab83396), are all their assay buffers the same? It would be quite difficult to divide the tumour sample to be lysed by the different assay buffers in each of the kits, so is that possible to use all 4 kits on the same tumour lysate?

Happy New Year 2012 to you all .. I had a discussion with my boss today regarding the kits that you have offered. I would like to know following things.. · I understand that the kit is not able to directly measure the enzyme activity. However, we can correlate it back to enzyme activity by measuring color change, which is acceptable.. I would like to know what is this color change resulting from?? (i.e.) I would like to know what is the reaction that is happening for this color change formation… I would be interested to measure this color change, only if it is happening due to product formation….(i.e.) I won’t like to measure, if this color change is due to substrate consumption… The only reason behind this as follows: o For example, if the reaction is: known substrate+co-factor, buffers etcsample (containing enzyme to be tested)product § Now at time T0, if the substrate concentration is 100uM (for example), then the product concentration is Zero. § At time T1, if substrate concentration drops 95 uM, the product formed will be 5 uM ideally.. and so on for T2, T3… However, if we are evaluating for 5% change only, then generally if the kit is working based on substrate consumption, it is difficult to measure this difference significantly. Whereas, if the kit is working based on color change due to product formation, then that zero to 5 uM amount is generally found significant…. Hence, I would like to know on what method your kits are working… Please let me know if I was clear in explanation.. .I can arrange for the skype meet if there are any queries.. May I know how long does it take for the delivery to Singapore for the kits…? Thanks and regards

Thanks for contacting me. I have looked at these kits that we discussed and here are my thoughts: Malate Assay Kit (ab83391): This kit will only measure malate present in the sample. The enzyme used to produce the oxidized form of malate, the form that is detected by the probe in the kit, is considered proprietary by our supplying lab so unfortunately I can't provide any information regarding the oxidized form of malate. Maltose Assay Kit (ab83388): This kit could be used to measure alpha-D-glucosidase activity by measuring the conversion of maltose to 2 glucose molecules. Amylase Assay Kit (ab102523): This kit could be used to measure alpha-amylase activity by measuring the specific cleavage of ethylidene-pNP-G7 substrate. Pyruvate Assay Kit (ab65342): This kit could be used to measure pyruvate oxidase activity by measuring the oxidation of pyruvate present in the reaction. PicoProbe™ Acetyl CoA Assay Kit (ab87546): This kit can only be used to measure free acetyl CoA present in biological samples. I hope this is helpful. There is no stipulation as to what kind of 96 well plates are used as long as the detection method to be used is compatible. For example, if choosing to measure fluorescence black plates are needed, or if measuring luninescence or absorbance, white plates or clear plates would be most appropriate respectively.
