  • General notes

    Cell line: Ramos (Burkitt's lymphoma).
    Growth media: RPMI 1640 medium with 2 mM L-glutamine adjusted to contain 1.5 g/L sodium bicarbonate, 4.5 g/L glucose, 10 mM HEPES, 1.0 mM sodium pyruvate, and 10% heat-inactivated FBS.

     

    Ramos cell lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylene diamine tetra acetic acid, 1 mM phenyl methyl sulfonyl flouride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecyl sulfate, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The cell lysate was boiled for 5 min in 1 x SDS sample buffer (0.045 M Tris-HCl pH 6.8, 10% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue), containing 0.05 M DTT.

  • Background

    Derived from a caucasian Burkitt's lymphoma which does not possess the Epstein Barr Virus (EBV) genome. EBV infectability and permanent conversion into EBV positive sub-lines is possible by in vitro infection. The cells have B lymphocyte characteristics, with surface associated mu and kappa chains. Cells are used as model of B lymphocytes and for apoptosis studies.

The Ramos (Human Burkitt's lymphoma cell line) Whole Cell Lysate (ab3955) should be stored as directed on the datasheet of this product:
"Upon delivery aliquot and store at -20°C or -80°C."
It is important to avoid freeze / thaw cycles with this product to maintain the quality. If stored as instructed we would expect the lysate should be stable for several months. We do not have exact expiration dates for this product. However, if you have a problem with the product within a 6 month period you would be covered by the Abpromise guarantee.

Inquiry: Dear Abcam, The inquiry type I have is not listed in your dropdown menu, I am indeed looking for a product, but hoping you can help with some product information regarding ab3955, Ramos cell lysate. It is of interest that you use this as a positive control for your anti-TLR4 antibody. We are looking into using Ramos cells in an assay, but there are indications this cell line does not respond to LPS, despite expressing TLR4. I am happy to confirm that Ramos cell do express TLR4 and can be used a positive control for TLR4.

I amnotaware that Ramoscell areunresponsiveto LPS.

Please consider thatI amnot specialised in this particular field and do not want to provide information that might be incorrect.

Therefore we encourage customers to consult the latest literature available through PubMed and other resources in order to find the most up-to-date information about their specific research interests.

Afterquick search,I could not find any data that Ramos cells are un-responsive to LPS in general. Furthermore I found publications were LPS is used frequently with Ramos cells. Here some examples:

Thnak you for your enquiry. I have performed a search of the literature and I have found no reference to the use of Ramos cells as a positive control for TLR7. For our own antibodies against TLR7 we make the recommendation of human spleen cells and A20 cell lysates. Alternatively I found reference to the use of a whole Raw cell lysate as a postive control; depending on whether your antibody reacts with the murine homolog. To this effect we we sell a lysate: ab7187 RAW 264.7 (Mouse leukaemic monocyte macrophage cell line) Whole Cell Lysate I hope this information helps, please do not hesitate to contact us if you need any more advice or information.
