      Exosomes are best defined as extracellular vesicles that are released from cells upon fusion of an intermediate endocytic compartment, the multivesicular body (MVB), with the plasma membrane. This liberates intraluminal vesicles (ILVs) into the extracellular milieu and the vesicles thereby released are what we know as exosomes. They contain abundant proteins, DNA, mRNA, and non-coding RNAs. They are now considered as an additional mechanism for intercellular communication, allowing cells to exchange proteins, lipids and genetic material. Increasing studies have shown that exosomes play an important role in tumour initiation, growth, progression, metastasis, drug resistance and immune escape.

      Fresh and lyophilized Exosome batches were analyzed and compared by WB in native conditions for exosomal markers, by anti-CD9 (Clone VJ1/20) and anti-CD63 (Clon TEA3/18) antibodies at a 1:1000 dilution (0.1 mg/ml).

    Quantification of Extracellular Vesicles by Single Molecule Array (SiMoA) Technology

    The purpose of our studies is to quantify the plasma levels of specific proteins considered breast cancer biomarkers such as extracellular vesicles (EVs) through liquid biopsy. To do this, we use the Single Molecule Array (SiMoA) technology, a new ultrasensitive digital ELISA immunoassay that can quantify very low concentrations of protein biomarkers present in human plasma without any previous purification process. For the construction of the calibration curve, we need EVs standards that express on their surface the markers commonly used to characterize EVs present in the biological fluids.
    In this regard, we wanted to test Abcam Lyophilized MCF7 Exosome Standards (Catalog Number: ab239691) to see if they express the markers of our interest (biomarkers belonging to the tetraspanin family) in order to obtain a calibration curve that allows a high detectable region.
    From the results obtained, it was possible to observe that Abcam lyophilized MCF7 exosomes standards express on their surface the markers specific for a subgroup of breast cancer EVs. Moreover, it is possible to obtain a calibration curve characterized by wide range allowing the quantification of the EVs levels adopting a very low sample dilution (only 1: 4).
    Healthy individuals used as a control group were considered for this analysis. Samples were analyzed in duplicate using 8 µl of plasma for each well obtaining an excellent reproducibility intra-plate (CV% < 10%).
    Considering the results obtained from our analysis we can attest that it is possible to use Abcam lyophilized MCF7 exosomes standards for the determination of EVs plasma levels in breast cancer patients by adopting very low dilutions allowing the use of few microliters of sample.

    In this abreview we haven't reported in detail all the phases of the essay and the antibodies used as it is our intention to publish the work.

    Repeatability Study using Multispectral Nanoparticle Tracking Analysis (m NTA)

    Lyophilized MCF7 Exosome (100 ug) sample was acquired from Abcam (ab239691). The sample was reconstituted according to protocol with XZero water to achieve a final exosome concentration of 1 µg/µL. After the addition of water, the vial was briefly vortexed to ensure homogenous distribution of particles. When preparing dilutions for measurement, 10 µL exosome sample was diluted serially using 0.02 μm filtered 20mM PBS at pH 7.4. The remaining reconstituted sample was stored at -20°C.

    Measurements were carried out at ambient laboratory temperature using the ViewSizer 3000 multispectral Nanoparticle Tracking Analyzer (m NTA). The ViewSizer 3000 features three simultaneous operating lasers with independently adjustable power and a color camera. The patented sample cuvette apparatus also allows stirring between videos, providing fresh and randomly sampled suspensions to be tracked and measured.

    The cleanliness of the cuvette was first verified with 400 µL of PBS buffer using the ViewSizer 3000. A sufficiently cleaned cell should yield no more than 10 particles in one video recording. Once verified, the buffer was pipetted out and replaced with 400 µL of Lyophilized MCF7 Exosome diluted 4,000 times with 0.02 μm filtered PBS buffer.

    Six replicates of Lyophilized MCF7 Exosome were measured over a period of 40 minutes. The exosome showed the main size distribution from 60 – 400 nm and concentrations averaging at 2.13E+11 exosome/mL. The repeatability of Lyophilized MCF7 Exosome concentration measurements over six replicates resulted in a CV of 5.80%. This is a remarkable mention as this indicates excellent instrument and sample stability. Unlike monodispersed latex standards, polydispersed biologic materials present a much greater challenge for accurate and precise measurements. Lyophilized MCF7 Exosome may be used to optimize parameters in NTA.

    Here we evaluated Lyophilized MCF7 Exosome as a preliminary study using the multispectral NTA, we will further study sample behavior for reproducibility and linearity.
