    FITC Conjugation Kit / FITC Labeling Kit ab102884 uses a simple and quick process for FITC labeling / conjugation of antibodies. It can also be used to conjugate other proteins or peptides. Learn about our antibody labeling kits and their advantages.


    We recommend FITC Conjugation Kit ab188285 as an alternative this kit using a faster, newer protocol.


    To conjugate an antibody to FITC using this kit:
    - add modifier to antibody and incubate for 3 hrs
    - add quencher and incubate for 30 mins
    The FITC conjugated antibody can be used immediately in WB, ELISA, IHC etc. No further purification is required and 100% of the antibody is recovered for use.

    1 Using the maximum amount of antibody may result in less labelling per antibody.

    2 Ideal antibody concentration is 1mg/ml. 0.5 - 1 mg/ml can be used if the maximum antibody volume is not exceeded. Antibodies > 2 mg/ml or < 0.5 mg/ml should be diluted /concentrated.

     

    Buffer Requirements for Conjugation

    Buffer should be pH 6.5-8.5.

    Compatible buffer constituents 
    If a concentration is shown, then the constituent should be no more than the concentration shown. If several constituents are close to the limit of acceptable concentration, then this can inhibit conjugation.

    50mM / 0.6% Tris1 0.1% BSA 50% glycerol
    0.1% sodium azide PBS Potassium phosphate
    Sodium chloride HEPES Sucrose
    Sodium citrate EDTA Trehalose

    1 Tris buffered saline is almost always ≤ 50 mM / 0.6%

    Arginine Glutathione DTT

    If a constituent of the buffer containing your antibody or protein is not listed above, please check the FAQ or contact us.

    Only purified antibodies are suitable for use, ie. where other proteins, peptides, or amino acids are not present: antibodies in ascites fluid, serum or hybridoma culture.

    Storing and handling conjugation kits

    Lyophilized Lightning-Link® components are hygroscopic.

    Kits are intentionally shipped at ambient temperature with silica gel to avoid exposure to moisture. Upon receipt, store the kit frozen and protect from moisture. Before opening the outer container, allow the lyophilized components to reach room temperature to minimize condensation.

    Lim, Tony KY, et al used FITC Conjugation Kit - Lightning-Link® (ab102884) as part of examining traumatic nerve injury. They used the kit to conjugate FITC to mouse anti-neurofilament 160/200  for use in immunohistochemistry.B, Contralateral nerves were stained for hypoxyprobe-1, DAPI, and neurofilament 160/200 (NF-M/L, axons). E, Ipsilateral nerves were subjected to the same staining. The area in proximity to the injury site is shown. Hypoxia, as shown by hypoxyprobe-1 staining, is observed in a region close to the injury site, in which axons, macrophages, and Schwann cells are all found to be hypoxic. Inquiry: I have purchased the antiphosphotidylserine antibody (ab18005), and I am wondering about a standard protocol to use this antibody along with the EasyLink FITC Conbjugation Kit (ab102884) and EasyLink HRP Conjugation Kit (ab102891) also purchased from abcam. Because I have noticed in the data sheet that the Anti-PS is suspended in 0.1M Tris gycine, 30% Glycerol, and 0.15M Sodium Chloride, and in the EasyLink data sheet, it is mentioned that buffers wirh primary amino acids and Tris-based should be avoided. So how can I go around to conjugate the Anti-PS antibody with the EasyLink FITC?

Is the quantity present in this antibody enough to be used with 2 EasyLink Conjugation kits, one with FITC and another with HRP? and how can I avoid thaw-freeze cycles to maintain the stability of the product if several phases of the experiment require multiple uses? Is there a way that the Ab can be resuspended in other vials and divided to be preserved and used upon need seperately?

Thank you for your inquiries.

I can confirm that additives in the buffer of the antibody will interfere with the labeling reaction.

We offer a kit that will concentrate the antibodyand also remove the additives:

We usually recommend to label 100ug of antibody with the kits you purchased to achieve an optimal result. This means that one vial of ab18005 is enough for only one conjugation reaction.

It is possible to only use 50ug, but this is not optimal.

After the labeling of the antibody we recommend to store it at 4C to avoid freeze /thaw cycles that will harm the conjugate-antibody-complex.

Sodium azide can be added, but only to conjugate that are NOT HRP. HRP enzyme activity is inhibited by sodium azide.

Thank you for contacting us.Ab69252 is batch tested using direct immunofluorescence in a microscopy assay (ICC/IF). This is a semi-quantitative assay, but we estimate approximately 20% variability between lots. This variability is due to the fact that ab69252 is a polyclonal antibody raised against a whole cell preparation. For a less batch variable product, I would recommend using one of our mousemonoclonal antibodies (ab23571 or ab128165)with our EasyLink FITC conjugation kit ab102884.

Our EasyLink conjugation kits allow for rapid and efficient conjugation of antibodies to a number of fluorophores. The antibody ab23571 is supplied at a low concentration, so it would need to be first prepared with the concentration kit ab102776. For more information, please see https://www.abcam.com/EasyLink

The EasyLink conjugations require antibody concentrations of at least 1mg/ml. Below that, the conjugation becomes inefficient and you risk wasting the conjugate, not to mention the antibody. For antibodies of lower concentrations, we recommend bringing them up to 1mg/ml or more with a concentration columns. We have two sizes: one column or three columns.

Thank you for your enquiry.

Adding the quencher ensures that the antibody and conjugate will no longer be reactive. This step does not help to remove the remaining free FITC molecules. They will remain in the solution but will be inactive. In most applications the remaining free FITC will be washed away during the first wash after incubation of the labeled primary antibody with sample of interest.

We can not provide credit note if customer is doing ELISA because it is not guaranteed. It is guaranteed for IHC and ICC/IF, flow cytometry or other applications that involves fluorescence.
