    FITC Conjugation Kit / FITC Labeling Kit ab102884 uses a simple and quick process for FITC labeling / conjugation of antibodies. It can also be used to conjugate other proteins or peptides. Learn about our antibody labeling kits and their advantages.


    We recommend FITC Conjugation Kit ab188285 as an alternative this kit using a faster, newer protocol.


    To conjugate an antibody to FITC using this kit:
    - add modifier to antibody and incubate for 3 hrs
    - add quencher and incubate for 30 mins
    The FITC conjugated antibody can be used immediately in WB, ELISA, IHC etc. No further purification is required and 100% of the antibody is recovered for use.

    1 Using the maximum amount of antibody may result in less labelling per antibody.

    2 Ideal antibody concentration is 1mg/ml. 0.5 - 1 mg/ml can be used if the maximum antibody volume is not exceeded. Antibodies > 2 mg/ml or < 0.5 mg/ml should be diluted /concentrated.

     

    Buffer Requirements for Conjugation

    Buffer should be pH 6.5-8.5.

    Compatible buffer constituents 
    If a concentration is shown, then the constituent should be no more than the concentration shown. If several constituents are close to the limit of acceptable concentration, then this can inhibit conjugation.

    Only purified antibodies are suitable for use, ie. where other proteins, peptides, or amino acids are not present: antibodies in ascites fluid, serum or hybridoma culture.

    Storing and handling conjugation kits

    Lyophilized Lightning-Link® components are hygroscopic.

    Kits are intentionally shipped at ambient temperature with silica gel to avoid exposure to moisture. Upon receipt, store the kit frozen and protect from moisture. Before opening the outer container, allow the lyophilized components to reach room temperature to minimize condensation.

Question

We are involved in a study that uses bone marrow stromal cells from goat (BMSC), and we’d need to characterize the cells, by means of flow citometry, against CD-34 and CD-90. Our difficulty is to find a specific antibody, that reacts positively with goat cells. I’ve searcher on several suppliers and only on yours I found a CD-34 with positive reactivity against sheep. Do you know if someone has used it against goat, as goat BMSC are being used on several studies?

Would you recommend this antibody: Anti-CD34 antibody [EP373Y] (ab81289), to be used against goat?

We also would like to mark the goat BMSCs cells against the CD-90. In your site I did not see a reference to positive reactivity against goat, sheep or cow. Do you know if anyone has used one of the listed antibodies for goat studies, and if they have shown positive reactivity?

In case of a positive answer which one would you recommend us?

In the case that abcam can supply us with this two antibodies, we would like them marked with fluorescent probes (not FICT or PE, as we’ll combine this antibodies with others we’ve bought a few months ago and that are marked with PE and FICT), which are the possibilities that you offer?

Do you have a dealer in Portugal? Frilabo or Alfagene?

We currently do not have any publication information regarding its use in sheep BMSCs. I am sorry we can't offer any tested data. This antibody however was tested with sheep lysates and gave good staining so it is more likely that the antibody with detect sheep CD34 target. The use of ab81289 (https://www.abcam.com/CD34-antibody-EP373Y-ab81289.html) comes with Abcam Abpromise guarantee which states we will refund your money if the antibody fails to detect the CD34 target.

I have stored the kit in -20 C from the arrival until the day I tried to use it. The vial with the lyophilized material was allowed to stand in RT for approximately 5 min before the antibody was added. The volume I used was 130 ul, and more can not really be used... I was pipetting the antibody for a few times without succeding with resuspending it... After a while, I pipetted a few times for, but it was still not possible. Unfortunately we have lost a lot of antibody which I guess can not be used. Is it possible to get a vial sent to us as compensation, it would be highly appreciated!

Answer

I have just received an answer from lab. The reason they have suggested is the loose vial cap which could have unable to stop the moisture getting in the vial. So we are happy to replace the vial. I am now working with our goods in and goods out team for processing the replacement vial.

Could you let me know if you have sufficient amount of Modifer and Quencher reagent that could be used with new replacement vial of FITC mix.

Hi!
I have used your product ab102884 to FITC-conjugate ab´s for FACS. I used 2 of the vials in the package i received from you the 2nd of december (lot number GR60869_1) immediately, but the third one has been saved and stored according to your instructions until now. When I now wanted to use it for labelling it was not possible to resuspend the lyophilized material. I can not find any information saying that the product can not be saved for some time before using it, and it has not been saved here for such a long time. Is it supposed to be used immediately, or was it something wrong with this product? I was at the same time labelling three other antibodies with a new fresch FITC-conjugation kit of the same kind, and they were perfectly fine!

- Could you provide details how the vials were stored? If stored properly the kit viability is guaranteed for 6 months.
- How much volume of antibody you have been using? Try higher volume e.g. 100ul
- We recommend leavening the lyophilized vial to warm for couple of minutes and then adding antibody in it. Redispensing the liquid 2-3 time using pipette with gentle shake will help.

Answer

Thank you for your inquiry. Indeed, antibodies as most (unmodified) proteins have a carboxy group (-COOH) at their C-terminus, as you indicated. They also have an amine group (-NH2) at their N-terminus. Furthermore, antibodies contain amino acids which have carboxy and amine groups in their side-chains, such as Arginine, Lysine, Glutamate or Aspartate. Therefore, the coupling reaction with FITC that usually takes place via free amine groups forming a stable thiourea bond (as for example with our EasyLink kits) is absolutely feasible for the anti-Enrofloxacin/Ciprofloxacin antibody ab21035. However, as the Goat polyclonal Secondary antibody ab97230 is already conjugated to HRP, I would not recommend to label this antibody with FITC. Thank you for your enquiry. I can confirm that the antibody concentration should not cause an issue. The Easylink conjugation kits should still work with this lower concentration, although it will be it at a slightly reduced efficiency. I can recommend to check the datasheet of the kit carefully for further details. For example, if your antibody contains thermasol, this compound will inhibit conjugation. Also, we would recommend not to concentrate the antibody if it contains BSA. Higher concentrations of BSA will affect the conjugation. The three antibodies you have suggested in your latest e-mail are of significant interest to both myself and my supervisor – to the point where we are thinking that this is worth trying. I would ask why you believe these antibodies will work in a Flow cytometry assay when Abcam have not tested these antibodies for this method?  I’m also interested in learning more of the animal model you yourself used for your PhD. You have mentioned an unusual animal model and in this e-mail that the sequence similarity was fairly low, and yet worked so I would be interested to learn more about the model you used, what you were looking for and of course which antibody you chose, although If was not an Abcam one I can understand that you may not want to divulge this particular piece of information!  I believe that we will attempt to look at CD107a using your already conjugated antibody ab67283. An already conjugated antibody will be easier to use, and I’m assuming Cy3 is equivalent to PE (in that they fluoresce at the same wavelengths) which will be suitable for any further experiments should it work. I apologize for the delay of mine! 1) There are no particular requirements for antibodies to work in flow cytometry, so the mentioned antibodies should give positive results using this technique. It is more important to optimise the protocol according to the target (in particular the stimulation for inducible targets etc). From a technical point of view, it is recommended to use directly labeled antibodies and avoid staining with a secondary antibody, as this may increase unspecific background staining (especially when using a secondary that has not been optimised for the analyzed, non-standard species). Therefore, in case you need to test the unconjugated antibodies, I would recommend labeling them before use. Dear xxxx,   Maybe I made you confused. I'm sorry about that.   1. What protein is she trying to detect? HLA with antibody ab31574? Our customer applied HLA A11 positive patients samples so they should show 100% of positive results. I have attached image of ab31574+ ab6650 result. 2. Which antibody was being purified for use with the FITC conjugation? She used ab33807 for conjugate FITC(ab102884). 3. What was the Anti-biotin antibody (FITC) (ab6650) being used for? She used this antibody with ab31574 and the result is attached. At the first time, she tried ab31574+ab6650(detection with biotin) but, the results were not satisfied so she tried FITC.   If you need more information, please let me know.

Thank you for clarifying those details. It may be that it is not the kits or the secondary antibody which is producing the undesired effects. It may be that the primary antibody binding may not be optimal. I am sorry to ask for even more information but I think it will help to find out where the problem may be and to try to get the experiment working. Firstly, could you clarify what each of the different colours stand for on the image you supplied. There is a green, red and pink population of cells as far as I can see. Also, if you could ask you customer to fill out the form that I have attached this may further identify any issues in the primary antiobody binding. We would not recommend using the protein purification kit (ab102784) with IgM antibidies, such as ab31574 as the kit relies on binding of the antibody to protein A resin. This interaction is very weak with IgM antibodies. The direct FITC conjugation of ab31574 may also not have been successful as the antibody used is already conjugated with biotin. I am sorry for any inconvenience caused to the customer. I will try to resolve this issue as soon as I can.

Thank you for your e-mail, I apologise for getting back to you so late, I have been away at a conference and then on holiday, and didn’t want to reply until I had a chance to think about your offer.  I think your offer is very generous, especially considering the lower than recommended sequence comparability. However this sadly will not help me with my goal. Abcam offers a plethora of different CD107a antibody products and also have more than one clone – Choosing the antibody most likely to work, and then paying over £200 for it is something my supervisor is unlikely to approve despite the potential benefits of this project.  I would appreciate your assistance is identifying the clone most likely to work as you mentioned in your e-mail – I found it most encouraging to hear that you have also worked on a similar project in the past, but would also like to ask if there is any other way we could go about attempting this project. Thank you very much for your message. As I myself have been in the very same position, I very well understand that you and your supervisor hesitate to test our different CD107a antibodies. At that time I had the permission by my supervisor to test various antibodies for their cross-reactivity - and was extremely happy to find out that the very first one I tried did work, despite an sequence similarity of only 68% across the full-length protein. Unfortunately, due to logistic reasons we cannot offer testing customers' samples. That is why we offer the testing discount to our customers. Also, I would like to apologize that I did not think earlier of the other possibility that you have: We do have three LAMP1-antibodies available whose immunogens share a 100% sequence similarity with cow. We have not specifically tested this yet, however, we of course expect them to cross-react. Although these antibodies have not been tested in flow cytometry, there is no reason why they should not work. Two of them are not fluorochrome-labeled, but this issue can easily be dealt with, as I am sure you know.
