    Overview

    • Product overview

Gold nanoparticles at Abcam have a proprietary surface coating that can withstand the most extreme conditions (e.g. 2.5 M NaOH at 70°C for 2 hours), making our Gold nanoparticles an ideal reagent for a wide range of applications.

Our Gold streptavidin conjugates share the stability attributes of the core nanoparticle. Furthermore, since the streptavidin is covalently bound to the surface coating of the gold nanoparticle it is unable to desorb, which is a major problem faced by competing products whereby the streptavidin is directly attached to the metal. Our Gold Streptavidin binds with high affinity to biotinylated molecules.

A common application for Streptavidin Gold is to be used as a detector in Lateral Flow Assays (LFAs).

      Notes:

      Schematic for ab186864

The 3 possibilities of binding are shown in the image:

A   Ideal concentration of Biotin conjugated Rabbit IgG (50-100 μg/ml) binds to the Streptavidin GOLD producing a strong signal on the test line (Goat anti-rabbit IgG).

B   Excess Biotin conjugated Rabbit IgG (>400 μg/ml) results in competition between the unbound Biotin conjugated Rabbit IgG and Streptavidin GOLD for the test line.

C   A lack of Biotin conjugated Rabbit IgG (<10 μg/ml) leads to a weak test line.

    Question

    1. What is the streptavidin concentration? Is it approximately 3mg/mL?

    2. Do you know approximately how many biotin sites are free for each streptavidin?

    3. Is there any reason (that you know of) that this reagent could NOT be used for mass cytometry (i.e. CyTOF)?

    Answer

    1. What is the streptavidin concentration? Is it approximately 3mg/mL?
We have never tested exactly how many streptavidin particles there are per gold nanoparticle however I can tell you the number of gold nanoparticles coated in streptavidin within the vial is 1.8 x10^11.

    2.
Unfortunately we don't know how many binding sites there are available on the Streptavidin because of the way they coat the surface of the gold nanoparticles, however it is worth noting that because the biotin-streptavidin bond is so strong it should only take one biotin to bind the streptavidin.

    3.
We have never tried using our GOLD-streptavidin conjugation reagent in mass cytometry. However if other similar products are used for this application, I am not aware of any reason why our product could not be used.
