    • Product overview

Abcam’s High-Density Lipoprotein (HDL) Human in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Human HDL in plasma, serum, milk, and cell culture supernatant.

A HDL specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently biotinylated HDL is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is inversely proportional to the amount of HDL captured in plate.

The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

    • Relevance

High-density lipoprotein (HDL) is one of the five major groups of lipoproteins, which, in order of sizes, largest to smallest, are chylomicrons, VLDL, IDL, LDL, and HDL, which enable lipids like cholesterol and triglycerides to be transported within the water-based bloodstream. In healthy individuals, about thirty percent of blood cholesterol is carried by HDL.

Blood tests typically report HDL-C level, i.e. the amount of cholesterol contained in HDL particles. It is often contrasted with low-density or LDL cholesterol or LDL-C. HDL particles are able to remove cholesterol from within artery atheroma and transport it back to the liver for excretion or re-utilization, which is the main reason why the cholesterol carried within HDL particles (HDL-C) is sometimes called "good cholesterol" (despite the fact that it is exactly the same as the cholesterol in LDL particles).

    Question

Inquiry: I would like to have more information on the capture antibody which is immobilized on the provided microplates of Abcam HDL ELISA kit. Particularly: - Is it monoclonal or polyclonal? - Is it specific for any apolipoprotein associated to HDL? - Does it show cross-reactivity with lipoprotein(a)?

    Answer

The ab125961 Human HDL ELISA Kit antibodies are polyclonal antibody against total HDL and recognize lipoprotein in the HDL.  It has some cross reactivity with Lp(a).
