    Human IL-8 ELISA kit is designed for the quantitative measurement of IL-8 supernatants, buffered solutions, serum and plasma samples. (Genecards: CXCL8)


    A monoclonal antibody specific for IL-8 has been coated onto the wells of the microtiter strips provided. Samples, including standards of known IL-8 concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for IL-8 are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of IL-8 present in the samples.


    This kit will recognize both endogenous and recombinant Human IL-8.

    IL-8 is a chemotactic factor that attracts neutrophils, basophils, and T-cells, but not monocytes. It is also involved in neutrophil activation. It is released from several cell types in response to an inflammatory stimulus. IL-8(6-77) has a 5-10-fold higher activity on neutrophil activation, IL-8(5-77) has increased activity on neutrophil activation and IL-8(7-77) has a higher affinity to receptors CXCR1 and CXCR2 as compared to IL-8(1-77), respectively.

    This kit was used to measure the level of IL-8 secretion in to the medium after infecting cells with E.coli strains. The kit was nicely sensitive to detect the level of IL-8 in the medium and was very helpful.
    The standards and the control vial worked very nicely and gave a nice curve.

    Pourriez-vous svp me dire, pour chaque référence, s’il est possible de modifier la gamme de standards.

    C’est-à-dire, pour votre kit IL8 le standard est reconstitué pour obtenir du 2.000 pg/mL, point le plus haut de la gamme.

    Est-il possible de le reconstituer dans un plus petit volume, pour obtenir par exemple du 4.000 pg/mL et ainsi pouvoir commencer la gamme à 4.000 ? à partir de quelle concentration le standard est-il soluble ?

    Et si oui, la qualité du test, la spécificité/sensibilité/précision du test est-elle tjs garantie ?

    Pour la MMP1, idem, est-il possible d’aller plus haut que 18 ng/mL ?

    Je vous demande cela car je souhaite analyser des surnageants de culture cellulaire, précisément de milieu de culture de traitement de tissus ex vivo. Généralement les concentrations en IL8 sont fortes, donc j’aurai besoin d’une gamme de standards qui aille le plus haut possible, pour ne pas avoir à diluer trop fortement mes surnageants et surtout que les valeurs ne soient pas toutes hors gamme.

    Nous ne recommandons pas de modifier les concentrations des courbes standards mais plutôt de diluer vos échantillons.

    Toutefois, si vous voulez vraiment changer les courbes standards ceci est possible. Cependant en augmentant les concentrations de la courbe standard, vous augmentez considérablement le risque de saturer l'anticorps qui peut conduire à un effet de plafonnement (une courbe non linéaire). N’oublions pas que l’anticorps de capture a aussi une limite du taux de protéine qu’il peut capturer.

    Veuillez noter que beaucoup de tests ont été réalisé pour déterminer les concentrations standards optimales pour les anticorps et donc si vous décidez de modifier les gammes de standards nous ne pourrons plus garantir que ces produits fonctionneront.

    De plus, nous ne savons pas à partir de quelle concentration les standards sont solubles.

    Pour ab100603, nous recommandons de reconstituer le standard dans 400 ul de diluant pour faire 100 ng/ml. Ceci est après dilué ˜5.555 fois pour faire une concentration de 18,000 pg/ml. Je vous conseille donc de diluer le standard de 100 ng/ml moins que 5.55 fois pour obtenir une gamme plus concentré.

    Pour ab46032, nous ne recommandons pas de reconstituer le standard dans un plus petit volume, pour obtenir 4.000 pg/mL.

    Thank you very much for your call yesterday and for your patience while I have been in touch with the lab about ab46032.

    This kit is suitable for all body fluids. Urine samples should be diluted in standard diluent buffer (containing PBS BSA). We have no data to report on urine samples, as the titers are variable, but we suggest starting the dilution at 1:1 (50 uL of samples and 50 uL of diluent).

    Thank you for contacting us. I apologise for the delay.
    Unfortunately any of these kits have been tested with human ascites. However, they should work as they are suitable for all body fluids.
    The only problem I would suspect is the high concentration of IgG in ascites which can interfere with the ELISA. I would suggest diluting samples 1:2 or 1:5 to minimize the matrix effect.

    Thank you for your phone call yesterday and for your questions about our ELISA kits. I have heard back from the lab regarding ab46032, and unfortunately, the kit was never assayed on murine samples. So, I would not recommend this kit to use with mouse samples, but you may be able to find a mouse IL8 kit from another vendor by searching on www.biocompare.com. Please let me know if you have any questions!

    Thank you for your enquiry. There's no Thimerosal in any component of the kit. However, we use azide (0.1%) in standard diluent and Biotinylated antibody diluent, and Proclin (0.1%) in HRP diluent.
