      Abcam’s MMP3 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MMP3 pro and active forms in serum, plasma, and cell culture supernatants. (Collect plasma using heparin as an anticoagulant. EDTA and Citrate are not recommended).


      This assay employs an antibody specific for Human MMP3 coated on a 96- well plate. Standards and samples are pipetted into the wells and MMP3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MMP3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

      Additional Notes: Incubation times are a little longer than in some other kits, which must be remembered when planning multiple experiments, but the data generated is of a better quality overall.

      The graph shows the differing levels of MMP3 released by primary fibroblasts exposed to several different active ingredients dissolved in media for several days prior to media collection and analysis.

      (Sorry I cant say what the actives actually are due to business nature of the work).

      Please see below customer's answers:

      1.) When has the kit been used and when have the components been reconstituted? How has the kit before and after reconstitution been stored?

      The kit was reconstituted and used on the same day, May 21st 2012 (together with the MMP-1 kit). Both before and after reconstitution they were kept at 4 deg C


      2.) Did you have any troubles reconstituting the standard?


      No. We subsequently tested the standard dilutions by dot blot with another anti-MMP3 antibody an obtained a reasonable signal.

      3.) Have you any comment on which component might be faulty? Have you, for example, tested whether the adding some of the HRP conjugated antibody to an aliquot of the substrate itself would give a signal?


      We have no idea for “ faulty component” but we can mention that the substrate was OK since we used the pooled substrates from kit MMP-3 and MMP-1 for both assays - in MMP1 plate we got a nice standard curve. We have indication that standard was OK since we got reaction with another batch of anti MMP3 antibody in a different method. We did not test the HRP antibody.


      4.) Just for our records, what kind of samples have you used?


      Our samples are skin surface washes of patients suffering from dermatitis and psoriasis (a small vessel with PBS is attached to skin for 30 min). Lesional skin from such patients is known to over-express these proteins. And indeed, in a previous group of patients tested by dot blot (and by using different antibodies) we obtained the expected results.

      T ELISA on MMP3 (ab100607) kit, strictly following all the instructions and using all the materials / solutions in proper dilutions as indicated in the kit. The only difference was that incubation with the substrate was prolonged to 45 min, after seeing very weak response in 30 min. However, the signal was still weak with a poor standard curve. The file with original data is attached. For comparison, the customer included the data obtained in the ELISA with the MMP1 kit as well (ab100604), that was run in parallel to the MMP3 test, which gave a nice standard curve. In order to test the system, they performed a dot blot with the same samples (standard + experimental samples) , however with another MMP-3 antibody, which worked nicely. The standard curve and samples were detected, so the protein was there... Please advise.

      We have not yet received any negative feedback and I am not able yet to tell where the problem might is coming from. I would therefore appreciate if you can confirm some further details:
      1.) When has the kit been used and when have the components been reconstituted? How has the kit before and after reconstitution been stored?
      2.) Did the customer had any troubles reconstituting the standard?
      3.) Has the customer any comment on which component might be faulty? Has he for example tested whether the adding some of the HRP conjugated antibody to an aliquot of the substrat itself would give a signal?

      Sorry for this late reply but we would be interested to test the MMP-3 kit (ab100607) in serum samples of monkeys and get our 100 % discount since this kit has only be validated for human samples.

      In addition, we are also interested to measure the IL23 cytokine(ab64708) in serum samples of monkeys. This kit is also only validated for human samples. The range of this kit is 156.2 pg/ml - 5000 pg/ml but the sensitivity is less than 20 pg/ml. So samples can be analyzed up to 20 pg/ml. Do you think it would be possible to get a discount for this kit as well?

      How many sample volume do we need for the MMP-3 kit and for the IL-23 kit? We only have 300ul -400ul of serum sample available.

      I wish to test 400 serum samples using your ELISA kits for: 1. Osteoprotegerin (Catalogue number ab100617) 2. MMP3 (Catalogue number ab100607) 3. DKK1 (Catalogue number ab100501)   I wonder if you could answer a few queries for me: 1. Please could you provide me with a 'Kit Insert' for each kit? - I was unable to find a link on your website. 2. Can I use these specific kits on an automated ELISA machine? - I would prefer not to test 400 samples individually. 3. As I would like to order 6 kits for each biomarker (thus 18 kits in total) would you be able to give me a more competitive rate than that stated onthe website? 4.
