    Abcam’s Tissue Factor (CD142) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Tissue Factor concentrations in plasma, serum, tissue extracts and cell culture supernatants.


    A Tissue Factor specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently a Tissue Factor specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Tissue Factor captured in plate.


    The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Testing crossreactivity in Rhesus EDTA plasma

The Tissue Factor ELISA, using the Abcam human ELISA kit, was performed by an experienced laboratory technician at an accredited university. The goal of the experiment was to analyze the linearity of dilutions of rhesus macaque EDTA-prepared plasma samples, but more specifically, to analyze the cross reactivity for rhesus macaque.

Overall, the kit, like other Abcam ELISA kits, was easy to follow, clear in terms of directions, and brief yet thorough. The laboratory technician had no problems understanding any of the protocol’s steps.

Our experiment consisted of using EDTA-prepared Rhesus Macaque plasma samples from N=3 M. mulatta. Samples were stored prior to thaw at -80 degrees Celsius. Plasma was diluted to several different dilutions, as well as the suggested dilution from the kit (1:2, 1:4, 1:8, and 1:16) in the given diluent. The rest of the assay was performed per the protocol’s instructions.

Absorbance measurements of the diluted samples were very comparable between duplicates however, there was a clear lack of linearity between all dilutions’ Tissue Factor value for each sample (see image).

The standard curve was clean with a good range of ODs (see image).

The technician also performed a spike recovery, and found that, between pure diluent with known standard and plasma with known standard, plasma masked the known standard’s concentration in detection using this assay. A spiked concentration of 50 pg/mL was added to pure plasma, then the sample was diluted the same as unspiked sample. Resultant values were the same as unspiked plasma, i.e. we could not detected the spiked protein in rhesus plasma.

Overall, we do not recommend this kit for detection of Tissue Factor in EDTA-prepared Rhesus macaque plasma samples.

My customer wants to do his research on cellular lysates with this test, but in the datasheet it is not written how much cells is needed for this test.

The number of cells used per sample with ab108903 Human Tissue Factor ELISA Kit (CD142) will very much depend on the amount of TF in the specific sample being tested.  If the expression level is low, you might want to try 1 million cell per sample (per measurement).  If level is expected to be high, you can use much less.   This will need to be individually optimized by customers for their own experiments.


I am sorry It is difficult to recommend how many cells to use per sample without further details of the samples being used. But I hope this suggestion will be helpful and a good starting point.
