    Abcam’s Mouse IL-10 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of IL-10 in serum, plasma and cell culture supernatants.


    This assay employs an antibody specific for mouse IL-10 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse IL-10 antibody is added. After washing away unbound biotinylated antibody, HRP conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-10 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Additional Notes: Tissue samples were homogenised in T-PER (Thermo) and protein concentration measured at 5mg/ml with a BCA Assay. These could be diluted 1:5 - 1:100 in the Assay Diluent and incubated overnight at 4oC and the results were measurable with the kit.

I can confirm that these three ELISA kits ab108870 ab100747 and ab100697 have been tested on mouse plasma and serum samples. (We would not recommend to test whole blood).

Thank you for your inquiry.

I can confirm that these kit are suitable for the use with serum samples that have been frozen.

I recommend to not to mix/compare samples thathave been frozen with samples that have not been frozen unless the impact of freezing is tested in a control experiment first.
