    Abcam’s TGF beta 1 Mouse in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for accurate quantitative measurement of Mouse TGF beta 1 concentrations in Cell culture supernatant, serum and plasma (EDTA, citrate).

    Mouse TGF beta 1 specific antibodies have been precoated onto 96-well plates. Standards and test samples are added to the wells and then incubated at room temperature. After washing, a Biotin-conjugated anti-Mouse TGF beta 1 detection antibody is added then incubated at room temperature. Following washing Streptavidin-HRP conjugate is added to each well, incubated at room temperature then again washed. TMB is added and then catalyzed by HRP to produce a blue color product that changes into yellow after the addition of an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Mouse TGF beta 1 captured on the plate

  • Function

    Multifunctional protein that controls proliferation, differentiation and other functions in many cell types. Many cells synthesize TGFB1 and have specific receptors for it. It positively and negatively regulates many other growth factors. It plays an important role in bone remodeling as it is a potent stimulator of osteoblastic bone formation, causing chemotaxis, proliferation and differentiation in committed osteoblasts.
  • Tissue specificity

    Highly expressed in bone. Abundantly expressed in articular cartilage and chondrocytes and is increased in osteoarthritis (OA). Co-localizes with ASPN in chondrocytes within OA lesions of articular cartilage.
  • Involvement in disease

    Defects in TGFB1 are the cause of Camurati-Engelmann disease (CE) [MIM:131300]; also known as progressive diaphyseal dysplasia 1 (DPD1). CE is an autosomal dominant disorder characterized by hyperostosis and sclerosis of the diaphyses of long bones. The disease typically presents in early childhood with pain, muscular weakness and waddling gait, and in some cases other features such as exophthalmos, facial paralysis, hearing difficulties and loss of vision.

    An ELISA was conducted to measure TGF beta with and without 5-ion GCR. The kit was utilized as manufacturer guidelines indicated and samples were diluted 1:10. The assay was quick and effective and will become a standard for our research team moving forward.

    Elisa for Osteoarthritis markers for Osteoblasts growth

    TGF beta 1 evaluation was performed on so we could see if TGF beta was being expressed in bone as a marker for remodeling using Osteoblasts.
    Our Samples included: With and without Radiation
    Assay procedure was performed according to manufacturer instructions. We had to dilute our samples according to our BCA results to what the manufacturer suggested. This ended up being 1:50 since the readings were so high. The Elisa kit is very straight forward, with the normal steps. Readings were performed at 450 nm. Photos show the results of standard curve and Test run for the kit.
    This kit very simple to use if you have a basic knowledge of an Elisa. Our lab will be using more of these

    The assay was used on wt mice vs db/db mice, which are obese and diabetic. The mice also suffer from chronic kidney damage. The graph shows the expected result, there is an upregulation in TGFb in db/db mice kidney tissue.

    Cross-reactivity with human TGF-b1

    As we plan to do TGF-b1 determinations from supernatants of human and mice co-cultured cells, we checked the ability of this Kit to distinguish both forms, human and rodent.

    We measured samples of DMEM, DMEM 10% and 0,5% FBS and DMEM + purified hTGF-b1 protein. Although samples with low FBS gives us a low value of TGF, samples with hTGF-b1 gives very strong signal.

    We consider that specificity of the kit is not promised, so we want a discount check for the value of the Kit. Also we want to notice that we buy two different Kit to be absolute sure that the Kit is the problem not our manipulation, so we claim for both Kits...



    Despite of the Kit is very easy to use, we consider that detection is a long process due to a lot steps of washing comparing to hTGFb1 detection Kit.

    I am interested in purchasing your ELISA kits – ab108792 and ab119557, to detect changes in mouse hippocampal tissue supernatants. I have not used these kits before and do not want to use half the plate to determine the optimum condition for tissue preparation and supernatant dilution for your detection range. I was wondering if you have any advice or any previous unpublished data on whether your kits are suitable for brain tissue supernatants and what dilution range you would recommend?

    ab108792: We have never tested brain tissue supernatants so I do not know if it would work. I cannot recommend the dilution range.

    ab119557: Again, we have not tested brain tissue with this kit but there is a publication where a FlowCytomix kit, which uses the same antibodies, was used for brain tissues. Therefore, I would assume that this kit would work on your samples however, I cannot guarantee this.

    We do have reports concerning tissue samples for ab119557 indicating that lysis buffers containing 0.5% TritonX-100 (detergent) had no deleterious effect, but we did not test this ourselves.


    We also have not tested the effect of protease and phosphatase inhibitors.

    We recommend, that - as a positive control- you spike in some of the standard at a known concentration in your buffer and see if it performs as expected.

    The lysis buffer only should be used a negative control.

    The TGF beta 1 Mouse ELISA Kit (with color giving dyes) (ab119557) is the only kit we currently have which can be used to detect TGF beta 1 in mouse samples. Regrettably we have not as yet tested this kit with tissue samples. It may be that the kit would be compatible with such samples but we would not be able toguarentee this.
