    Abcam’s TNF alpha Rat ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat TNF alpha in serum, plasma, and cell culture supernatants.

    This assay employs an antibody specific for Rat TNF alpha coated on a 96-well plate. Standards and samples are pipetted into the wells and TNF alpha present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat TNF alpha antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TNF alpha bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm.

    Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation.
  • Involvement in disease

    Genetic variations in TNF are a cause of susceptibility psoriatic arthritis (PSORAS) [MIM:607507]. PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoidlike pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; arthritis of the sacroiliac joints and spine (psoriatic spondylitis).

    O-glycosylated; glycans contain galactose, N-acetylgalactosamine and N-acetylneuraminic acid.

    Question

    J&apos;ai testé des échantillons en parallèle d&apos;un autre de
    vos kit Ab100767 anti IL1.
    Les résultats pour l&apos;IL1 sont parfaits et corrèlent avec la littérature
    avec des CV <8%.
    Par contre la plaque de TNFalpha ab100784 (TNFalpha rat ELISA kit,Lot
    n°xx) donne des résultats totalement incohérents pour chacun des
    duplicates avec des CV de 20 à 110%.
    Etant donné que les echantillons ont été testés en parallel, le probleme
    semble venir que de la plaque (problème de coating?). Avez-vous eu des
    retours pour le lot Lot n°xx.
    il me reste 4 strips et le reste du kit si vous voulez faire des tests
    de votre coté.
    Je vous joint en doc attachés les résultats des deux kits pour que vous
    puissiez vous faire une idée

    Je suis désolée que vous avez eu ce problème avec ce kit. Nous avons pas reçu aucun autre feedback négatif, ni sommes nous au courant d'un éventuel problème avec ce lot. Comme vous avezpar contre fait le contrôle avec l'autre kit en parallèle, il semble clair que il y avait un problème avec le kit. Nous allons donc vous le remplacer. Je tiens juste quand-même à vous rappeler que même si le kit devrait être stable pour plusieurs mois à 4°C, notre garantie est normalement de 6 mois. Merci pour votre compréhension.

    Vous ne devez pas nous renvoyer les 4 strips. Je vais mettre en place l'envoie d'un kit de remplacement gratuit de ab100784. Nous allons l'envoyer à la même adresse que le kit original. Le numéro de commande est le xx et vous recevrez sous peu une confirmation de la commande. Veuillez s'il vous plaît ne pas hésiter àme contacter - de préférence aujourd'hui -si nous devons changer l'adresse de livraison ou si vous souhaitez un remboursement à la place du kit de remplacement.
