    Immunocytochemistry/ Immunofluorescence analysis of B16-F0 ( mouse melanoma epithelial cell-like) cells labeling SOX10 with purified ab227680 at 1:25 (3 µg/ml). Cells were fixed in 4% paraformaldehyde and permeabilized with 0.1% Triton X-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1:200 (2.5 µg/ml). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1:1000 (2 µg/ml) dilution. DAPI nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

    Immunohistochemical staining of paraffin embedded human cerebral cortex with purified ab109186 at a working dilution of 1/100. The secondary antibody used is ab97051, a HRP-conjugated goat anti-rabbit IgG (H+L), at a dilution of 1/500. The sample is counter-stained with hematoxylin. Antigen retrieval was perfomed using Tris-EDTA buffer, pH 9.0. PBS was used instead of the primary antibody as the negative control, and is shown in the inset.

Really interesting kit for oligodendrocyte antibodies initial testing

The combination of antibodies cover the major range of oligodendrocyte cells and is really interesting for running an initial test and decide which mark could be of higher interest.

Following the recommended dilutions provided in their individual datasheets the staining worked with no major problems and the specificity seems to be quite high.

The major inconvinient is that all antibodies are generated in rabbit, so that co-staining with the different antibodies in the same sample is not feasible. It could be more interesting to have at least pairs of mouse/rabbit antibodies which could make the kit more versatile and could help on the decission of combinations of them.
