Mature Oligodendrocyte Marker (Olig2, MBP, MOG, SOX10) Antibody Sampler Panel - Human, Mouse (ab254026)

Immunocytochemistry/ Immunofluorescence analysis of B16-F0 ( mouse melanoma epithelial cell-like) cells labeling SOX10 with purified ab227680 at 1:25 (3 µg/ml). Cells were fixed in 4% paraformaldehyde and permeabilized with 0.1% Triton X-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1:200 (2.5 µg/ml). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1:1000 (2 µg/ml) dilution. DAPI nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

Immunohistochemical staining of paraffin embedded human cerebral cortex with purified ab109186 at a working dilution of 1/100. The secondary antibody used is ab97051, a HRP-conjugated goat anti-rabbit IgG (H+L), at a dilution of 1/500. The sample is counter-stained with hematoxylin. Antigen retrieval was perfomed using Tris-EDTA buffer, pH 9.0. PBS was used instead of the primary antibody as the negative control, and is shown in the inset.

Immunohistochemical analysis of 4% paraformaldehyde-fixed, 0.2% Triton X-100 permeabilized frozen mouse brain (cerebrum) tissue labeling Myelin Basic Protein with ab218011 at 1/5000 dilution, followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor^® 488) (ab150077) at 1/1000 dilution (green). Positive staining in the axonal fiber tracts on rat cerebrum tissue section (PMID: 23144976) is observed. The nuclear counterstain is DAPI (blue).

Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (Alexa Fluor^® 488) (ab150077) at 1/1000 dilution.

Perform heat-mediated antigen retrieval by using ab94681 (Tris-EDTA buffer, pH 9.0).

Immunohistochemical analysis of paraffin-embedded human cerebrum tissue labeling Myelin Basic Protein with ab218011 at 1/5000 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) Ready to use. Cytoplasmic staining in human cerebrum (PMID: 22496821) is observed. Counter stained with Hematoxylin.

Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (HRP) Ready to use.

Heat mediated antigen retrieval was performed using Perform heat mediated antigen retrieval using ab93684 (Tris/EDTA buffer, pH 9.0). Tissue was counterstained with Hematoxylin. ImmunoHistoProbe one step HRP Polymer (ready to use). PBS instead of the primary antibody was used as the negative control.

Immunofluorescent staining of A-375 (human malignant melanoma cell line) cells labeling SOX10 with ab155279 at a 1/250 dilution.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of Human melanoma tissue sections labeling SOX10 with ab227680 at 1/100 dilution (0.97 ug/ml). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 6.0, epitope retrieval solution 1) for 10mins. Rabbit specific IHC polymer detection kit HRP/DAB (ab209101) was used as the secondary antibody. Hematoxylin was used as a counterstain. Mainly nuclear staining on the human melanoma, performed on a Leica Biosystems BOND™ RX instrument.

The section was incubated with ab227680 for 10 mins at room temperature.

Nuclear staining on the mouse breast, performed on a Leica Biosystems BOND RX instrument.
