  • Function

    May have a role in checkpoint signaling during mitosis. Enhances TP53-mediated transcriptional activation. Plays a role in the response to DNA damage.
  • Involvement in disease

    Note=A chromosomal aberration involving TP53BP1 is found in a form of myeloproliferative disorder chronic with eosinophilia. Translocation t(5;15)(q33;q22) with PDGFRB creating a TP53BP1-PDGFRB fusion protein.
  • Sequence similarities

    Contains 2 BRCT domains.
  • Post-translational
    modifications

    Asymmetrically dimethylated on Arg residues by PRMT1. Methylation is required for DNA binding.
    Phosphorylated at basal level in the absence of DNA damage. Hyper-phosphorylated in an ATM-dependent manner in response to DNA damage induced by ionizing radiation. Hyper-phosphorylated in an ATR-dependent manner in response to DNA damage induced by UV irradiation.
  • Cellular localization

    Nucleus. Chromosome > centromere > kinetochore. Associated with kinetochores. Both nuclear and cytoplasmic in some cells. Recruited to sites of DNA damage, such as double stand breaks. Methylation of histone H4 at 'Lys-20' is required for efficient localization to double strand breaks.

    Question

    The samples are primary cortical neuron cells from Fisher 344 rat (embryo of 18 days)

    left to grow for 6days and after adding the treatment the proteins were collected from the cell culture plate by lysis buffer (20mM tris-Hcl, 150mM Nacl, 1mMNa2EDTA, 1mM EGTA, 1% triton, 2.5mMsodium pyrophosphate 1mM beta glycerophosphate 1mMNA3VO4 ans 1ug/ul leupeptin ans 1mM PMSF)

    Protein conc were measured by bradford assay

    Answer

    Thank you for getting back to me and for providing some further details of the experiments. There may be several factors which contribute to lower band on the blot.

    Since proteinase inhibitors have been added to the lysis buffer, the chance of protein degradation is very minimal.

    1) Cross-reactivity: First of all it is important to mention that ab36823 has only been tested in human and mouse samples. No cross-reactivity information available regarding rat so we can't guarantee this antibody to work in rat samples. Although, it may work absolutely fine but we do not have any lab data to support it.

    2) Primary cell culture: Cells tend to dedifferentiate quickly in primary culture. So it is very important to apply good positive control with the samples and to interpret the results with caution. Do you have any preliminary data indicating the expression levels of 53BP1 in normal brain cells?

    3) Positive control: It may be worth running good positive control alongside with the samples to see if the band appears at the same size or not. For this purpose, I would perhaps recommend using any of these cell lines i.e. U205, HeLa, 293T, MO59K cell lysates (human) and MEF lysates (mouse).

    4) Isoforms: According to Swiss-Prot website, several isoforms and splice variants have been described and it may well be that in rat there are also different variants of 53BP1. Please check these sites for further information about the different forms:
