  • Function

    This is one of the several different receptors for 5-hydroxytryptamine (serotonin), a biogenic hormone that functions as a neurotransmitter, a hormone, and a mitogen. The activity of this receptor is mediated by G proteins that inhibit adenylate cyclase activity.

    ab85615 staining 5HT1A Receptor in primary hippocampal rat neurons/glia, (obtained from Neuromics, cat. no. PC35101), DIV14. The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% PBS-Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at 4°C with ab85615 at 5µg/ml and ab7291, Mouse monoclonal [DM1A] to alpha Tubulin - Loading Control. Cells were then incubated with ab150081, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 488), pre-adsorbed at 1/1000 dilution (shown in green) and ab150120, Goat polyclonal Secondary Antibody to Mouse IgG - H&L (Alexa Fluor® 594), pre-adsorbed at 1/1000 dilution (shown in pseudocolour red). Nuclear DNA was labelled with DAPI (shown in blue).

    Image was acquired with a high-content analyser (Operetta CLS, Perkin Elmer) and a maximum intensity projection of confocal sections is shown.

    IHC image of 5HT1A Receptor staining in Human Hippocampus formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab85615, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    Immunocytochemistry staining of 5HT1A Receptor using ab85615 in ioNEURONS/glut cells (Human iPSC-Derived Glutamatergic Neurons, ab259259), which were differentiated for 11 day post induction. The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% PBS-Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. Question

I am working on honey bee and interested in antibodies against Tyramine receptors. It might be that these antibodies ab85570,ab65833,ab16028, ab44635,ab60359, ab13896, ab85615, ab85937, ab102706, ab101913, ab101770, ab101914, ab102700, ab102701, might have raised against the peptide sequences that also recognize the AmTyr receptors. Answer

I am waiting for replies from some of the laboratories that produced these antibodies.

Of the antibodies for which we know the immunogen sequence (others are considered proprietary by the laboratories that produce the antibodies), the only two that have immunogen sequences with greater than 50% identity with the honey bee sequence you sent are:

ab85615, anti 5HT1A receptor, 77% (10/13 identical amino acids) and

ab60359, anti- 5HT4 receptor, 52% (22/42 identical amino acids).
