Function
Cleaves the membrane-bound precursor of TNF-alpha to its mature soluble form. Responsible for the proteolytical release of soluble JAM3 from endothelial cells surface. Responsible for the proteolytic release of several other cell-surface proteins, including p75 TNF-receptor, interleukin 1 receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor, MUC1 and the amyloid precursor protein. Also involved in the activation of Notch pathway. -
Tissue specificity
Ubiquitously expressed. Expressed at highest levels in adult heart, placenta, skeletal muscle, pancreas, spleen, thymus, prostate, testes, ovary and small intestine, and in fetal brain, lung, liver and kidney. -

Contains 1 disintegrin domain.
Contains 1 peptidase M12B domain. -
Domain
Must be membrane anchored to cleave the different substrates. The cytoplasmic domain is not required for the this activity. Only the catalytic domain is essential to shed TNF and p75 TNFR.
The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme. -
Post-translational
modificationsThe precursor is cleaved by a furin endopeptidase.
Phosphorylated. Stimulation by growth factor or phorbol 12-myristate 13-acetate induces phosphorylation of Ser-819 but decreases phosphorylation of Ser-791. -

Flow cytometric analysis of Wild-type HCT116 (human colorectal carcinoma epithelial cell, Right) / ADAM17 knockout HCT116 (Left) cells labeling ADAM17 with AB307269 at 1/500 dilution (0.1ug) (Red) compared with a Rabbit monoclonal IgG (ab172730) (Black) isotype control and an unlabeled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) at 1/2000 dilution was used as the secondary antibody. Gated on viable cells.
Positive staining on HCT116 cells (ab266873), while no staining on ADAM17 knockout HCT116 cells (ab255451).

Flow cytometric analysis of human peripheral blood mononuclear cell (PBMC) cells labeling ADAM17 with AB307269 at 1/50 dilution (1ug)/ Right compared with a Rabbit monoclonal IgG (ab172730) / Left isotype control.
Cells were stained with rabbit IgG or ab307269. Then stained with anti-CD14 conjugated to BV510.
Secondary antibody is Goat Anti-Rabbit IgG (Alexa Fluor® 488, ab150081) used at 1/2000 dilution.
Gated on viable cells.
