    Target

    • Relevance

      The non enzymatic reaction of reducing carbohydrates with lysine side chains and N terminal amino groups of macromolecules (amino acids, proteins, phospholipids and nucleic acids) is called the Maillard reaction or glycation. The latter products of this process, termed advanced glycation end products (AGEs), adversely affect the functional properties of proteins, lipids and DNA. In long lived tissue proteins, these chemical modifications accumulate with age and may contribute to the pathophysiology of ageing and long term complications of diabetes, atherosclerosis and renal failure.

      Immunocytochemistry/ Immunofluorescence - Anti-AGE antibody (ab23722)
      ICC/IF image of ab23722 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab23722, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

    Answer

    We have not tested the cross reactivity with ab23722 and the other types of AGEs that you mentioned. Only BSA-AGE and HSA-AGE have been tested, and the antibody shoes minimial cross reactivity (< 1%) with BSA and HSA alone.

    This antibody will detect endogenous and artificial AGEs. Mainly in certain diseases as diabetes, arteriosclerosis and Modbus Alzheimer AGE has been detected in various tissues (eye, kidney, artery, brain etc.). In principle all proteins can be affected, not only albumins.

    Inquiry: Hi, We had purchased ab23722 (anti-AGE antibody) awhile ago and I have come across some problems. Following a standard glycation protocol, we can detect the sample with a new anti-CML antibody (Ab125145), but not the Anti-AGE antibody. I am curious as to how the AGE modified BSA and HSA were prepared to create the immunogen to produce ab23722? As there are dozens of different AGE modifications possible it is of great interest to our group to know the specific modification detected by the anti-AGE antibody and whether the anti-CML and anti-AGE should be targeting similar structures.

    The immunogen for ab23722 was prepared as follows:
    Under sterile conditions a mixture of BSA and HSA was incubated with D-Glucose in phosphate buffer pH 7.4 at 37°C for some weeks. Low-molecular weight products and D-Glucose were eliminated by chromatography and dialyses against PBS pH 7.4. This final product was used for immunization of rabbits.

    We have purchased some antibodies (Anti-RAGE: ab37647 and Anti-AGE: ab23722) from Abcam and require some technical support in regards to one of them. We are looking to perform a Western blot using the anti-AGE antibody and noticed on the datasheet that there is some cross-reactivity between the antibody and BSA/HSA <1%. Does this mean that the probablility of cross reactivity is <1% or that there is cross reactivity present if the BSA/HSA concentration is <1%. If there is cross reactivity with BSA would you happen to have any recommendations as to what it would be appropriate to dilute this antibody in? I look forward to your response.

    Regarding ab62623, Stressed tissue such as Alzheimer's affected neurons. or Human brain pre treated with 10ug proteinase K for 40mins at 37oC are suitable positive controls.

    Regarding ab23722, human colon tissue was tested successfully.
