    Ubiquitinated; undergoes both 'Lys-48'- and 'Lys-63'-linked polyubiquitination. TRAF6-induced 'Lys-63'-linked AKT2 ubiquitination. When fully phosphorylated and translocated into the nucleus, undergoes 'Lys-48'-polyubiquitination catalyzed by TTC3, leading to its degradation by the proteasome.

    Lane 1: Wild-type HAP1 cell lysate (20 µg)
    Lane 2: AKT2 knockout HAP1 cell lysate (20 µg)
    Lanes 1 and 2: Merged signal (red and green). Green - ab131168 observed at 55 kDa. Red - loading control, ab8226, observed at 42 kDa.

    ab131168 was shown to recognize AKT2 when AKT2 knockout samples were used, along with additional cross-reactive bands. Wild-type and AKT2 knockout samples were subjected to SDS-PAGE. ab131168 and ab8226 (loading control to beta Actin) were both diluted 1/1000 and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1/10000 dilution for 1 h at room temperature before imaging.
