    Target

    • Function

Mitochondrial membrane ATP synthase (F(1)F(0) ATP synthase or Complex V) produces ATP from ADP in the presence of a proton gradient across the membrane which is generated by electron transport complexes of the respiratory chain. F-type ATPases consist of two structural domains, F(1) - containing the extramembraneous catalytic core, and F(0) - containing the membrane proton channel, linked together by a central stalk and a peripheral stalk. During catalysis, ATP synthesis in the catalytic domain of F(1) is coupled via a rotary mechanism of the central stalk subunits to proton translocation. Subunits alpha and beta form the catalytic core in F(1). Rotation of the central stalk against the surrounding alpha(3)beta(3) subunits leads to hydrolysis of ATP in three separate catalytic sites on the beta subunits.
    • Sequence similarities

Belongs to the ATPase alpha/beta chains family.
    • Cellular localization

Mitochondrion. Mitochondrion inner membrane. Peripheral membrane protein.

    Question

We are interested in an anti-AtpB antibody to detect AtpB of the bacterium Acinetobacter baumannii. We see that you sell an Anti-ATPB antibody [7E3F2] (ab110280) that recognizes Escherichia coli AtpB. Do you know if it reacts with Acinetobacter baumannii AtpB? If you do not know, we were wondering if you might be able to provide us with a small sample in order to determine if it reacts with Acinetobacter baumannii AtpB. If it does, we intend to purchase this antibody in the future; however, we would like to avoid spending the money on an antibody that may not work for us. If you would like, we can let you know if it does work for your product reference sheet. We would be very appreciative of your help.

    Answer

Thank you very much for your interest in ab110280.

To our knowledge, ab110280 has not been tested against samples of Acinetobacter baumannii . Therefore, I can offer a discount off a future purchase if you buy ab110280 now, test it in Acinetobacter baumannii, and submit feedback to us in the form of an Abreview. It doesn’t matter whether the Abreview is positive or negative, we would just really like to receive your feedback. The discount would be to the value of one free primary antibody.

If you are interested in this offer, please follow these steps:

    1. Reply to this e-mail to let me know that you would like to proceed and test ab110280 with Acinetobacter baumannii. I will then send a discount code. This code must be issued before purchasing ab110280 so please wait for my reply before ordering.

    2. Purchase ab110280 either by phone, fax, or online (www.abcam.com).

    3. Test it against Acinetobacter baumannii.

    4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews.

    5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any primary antibody ordered and the discount code is valid for 4 months after issue.
Please remember that submission of the Abreview is sufficient for the discount code to become active.

We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if ab110280 turns out to be unsuitable, you will still receive the discount on your next purchase after your Abreview has been submitted.

Please let me know if you have any questions about this offer and I would be happy to help you further.

    Question

Thank you very much for the prompt response. I wonder of you also have human mitochondria similar to the bovine preparation? Also, please let me know if the anti ATP synthase that you sell (particularly the mouse monoclonal ab5432 and the monoclonal antobody clone 7E3F2 - AB110280) interact with the bovine mitochondria - AB110338. I thank you in advance for your answer as soon as you can.

    Answer

Thank you for your email. Yes, we do carry human mitochondrial lysates as well: Human Liver Mitochondrial lysate (ab110339) Human Heart Mitochondrial lysate (ab110337) However, both lysates come in WB loading buffer (ready to be used in WB) and not iso-osmotic buffer as the bovine lysate. Regarding the antibodies: ab5432: This antibody has not been tested with bovine samples yet, but the sequence homology between rat (immunogen) and bovine is 96%. If you would like to try the antibody in bovine, you can take advantage of our testing discount program. For details please see below. ab110280: This antibody has been tested with bovine samples in WB and would be guaranteed for this species and application. Please see lane 2 for the WB data with bovine mitochondria. *Testing discount program: Here is some information about how the testing discount program works: The testing discount program is for customers who like to use an antibody/protein on an untested species/application. You would purchase the antibody at full price, test it and submit an Abreview with your data (positive or negative). On your next order you will receive a discount for ONE antibody at the full price (100%) of the antibody you have tested. The terms and conditions applicable to this offer can be found here: www.abcam.com/collaborationdiscount. *Abpromise guarantee: All our products are guaranteed for the tested species and applications as listed on the datasheet for 6 months after the purchase. For more details about our Abpromise guarantee please check this link: https://www.abcam.com/abpromise. I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
