Target

  • Function

    Component of the class I major histocompatibility complex (MHC). Involved in the presentation of peptide antigens to the immune system.
  • Involvement in disease

    Defects in B2M are the cause of hypercatabolic hypoproteinemia (HYCATHYP) [MIM:241600]. Affected individuals show marked reduction in serum concentrations of immunoglobulin and albumin, probably due to rapid degradation.
    Note=Beta-2-microglobulin may adopt the fibrillar configuration of amyloid in certain pathologic states. The capacity to assemble into amyloid fibrils is concentration dependent. Persistently high beta(2)-microglobulin serum levels lead to amyloidosis in patients on long-term hemodialysis.

In Western blot, ab75853 was shown to bind specifically to beta 2 Microglobulin. A band was observed at 12 kDa in wild-type HEK-293T cell lysates with no signal observed at this size in B2M knockout cell line ab266828 (knockout cell lysate ab256845). To generate this image, wild-type and B2M knockout HEK-293T cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in fluorescent western blot (TBS-based) blocking solution before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged.Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

     ab75853 Anti-beta 2 Microglobulin antibody [EP2978Y] was shown to specifically react with beta 2 Microglobulin in wild-type HepG2 cells. Loss of signal was observed when knockout cell line ab262325 (knockout cell lysate ab256846) was used. Wild-type and beta 2 Microglobulin knockout samples were subjected to SDS-PAGE. ab75853 and Anti-GAPDH antibody [6C5] - Loading Control (ab8245) were incubated overnight at 4°C at 1 in 1000 dilution and 1 in 20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

    Intracellular Flow Cytometry analysis of HeLa (human cervix adenocarcinoma) cells labelling beta 2 Microglobulin with purified ab75853 at 1/20 (red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. An Alexa Fluor® 488-conjugated goat anti-rabbit IgG (1/2000) was used as the secondary antibody. Black - Isotype control, rabbit monoclonal IgG. Blue - Unlabelled control, cells without incubation with primary and secondary antibodies.

Cells were fixed with 100% methanol and permeabilized with 0.1% Triton X-100. ab150077, an Alexa Fluor® 488-conjugated goat anti-rabbit IgG (1/1000) was used as the secondary antibody. DAPI (blue) was used as the nuclear counterstain. ab7291, a mouse anti-tubulin (1/1000) and ab150120, an Alexa Fluor® 594-conjugated goat anti-mouse IgG (1/1000) were also used.

    Control 1: primary antibody (1/100) and secondary antibody, ab150120, an Alexa Fluor® 594-conjugated goat anti-mouse IgG (1/1000).

Question

Hello, 
I am using the anti-beta 2 microglobulin antibody [EP2978Y] (ab75853) to detect human, mouse, and rabbit B2M expression in transfected cells.  The antibody works fine for human and mouse but does not cross react with rabbit.  I was just curious if you could tell me the exact sequence of the peptide immunogen used to raise this antibody against B2m?  Also, I did not see an antibody specifically stated to react with rabbit B2m on the website.  Do you happen to know if any other of your anti-B2M antibodies react with rabbit B2m.

In review of the immunogen sequence used for creating ab75853 I see this sequence share only 69.2% identity with rabbit beta-2-microglobulin. Generally we do not expect reactivity below 80%. Overall I have found that the sequence homologies of human and rabbit beta-2-microglobulin to be only 60.5% so I would not be able to recommend those items which have been created using purified protein or full length proteins and have not yet been epitope mapped. I have found one antibody on our catalog which has an immunogen with 83% identity against rabbit. This item, ab98327, has been created using a peptide corresponding to the N terminal region of beta-2-microglobulin-antibody and has been tested in western blot. To our knowledge, this product has not been tested in rabbit.

To whom it may concern,
I am interested in purchasing your primary B2M antibody catalog # ab75853 to stain some mouse cells for Flow cytometry expression of Beta 2 microglobulin. However, I am unsure of what 2nd antibody to get as well to read on the flow machine. I am looking for something that is excited by a red laser (640nm) as my cell will already have FITC and PE stain on them. Can you recommend a catalog number that fits this description for an anit-Rabbit secondary?

We plan on trying out one of your suggested anti-beta 2 microglobulin antibody (ab75853) first and participate in the testing discount program.   I just want to verify if there is any code associated with the program (is it the alphanumeric one in the brackets in the subject heading of the email?) and whether we need to notify you about which the products we plan to test in advance.  We will also most likely test the suggested ab99977 ELISA kit at some point but for now will order just the antibody listed above.

According to my information the mouse strain DBA/2NCrl has the haplotype H2d. Please encourage your customer strongly to double check this for the strain they mentioned (DBA II N strain).

We do not have any anti MHC class I antibodies that a re tested in IHC-P, but I could offer your customer to take part in the testing discount program for ab64368 that should be suitable for haplotype H2d.
